Objectives: To discuss the efficacy of multi-b value diffusion-weighted imaging(DWI) in evaluating non-invasive effect of endostar to the human colon carcinoma xenograft tumor in nude mice. Methods: HT-29 human colon cancer cells were seeded in 48 nude mice, the 48 mice were randomly divided into two groups, the control group and the experimental group each 24 mice, multi-b value diffusion-w-eighted imaging was acquired at prodrugs treatment and after treatment 2 days, 7 days and 14 days, observed the variation of apparent diffusion coefficient(ADC) v-ariation, eight nudes were sacrificed after scanning after treatment 2 days, 7 days,14 days and pathology immunohistochemical examination were done. Results: After treatment, ADC10 b, ADChigh, values of the endostar group tumor gradually increase, ADClow, ADCperf values of the endostar group tumor gradually decrease, four groups at different time points before and after treatment were compared was statistic-ally significant(F=29.986, 23.742, 21.598, 63.973, P<0.05), the endostar group pairwise comparison, before and after treatment 2 days, 7 days and 14 days the tumor observed index of ADC10 b, ADChigh, ADCperf was statistically significant, except after treatment 2 days and 7 days the tumor observed index of ADClow was no statistically difference.the other pairwise comparison the tumor observed index was statistically significant. After treatment, ADC10 b, ADChigh values of the control g-roup tumor gradually decrease, ADClow, ADCperf values of the control group tumor gradually increase, four groups at different time points before and after treatment were compared was statistically significant(F=34.501, 34.683, 17.144, 48.129, P<0.05), the control group pairwise comparison, before and after treatment 2 days, 7 days and 14 days the tumor observed index of ADC10 b, ADChigh, ADClow, ADCperf was statistically significant. There were no statistically difference between the two groups before treatments in 4 groups, and two groups after 2 days, 7 days, 14 day s the four groups observed indicators were statistically significant(P<0.05). After the endostar group and control group, the ADCperf result was positively correlated with MVD at three time points(P<0.05) and the ADC10 b result was negatively co-rrelated with MVD(P<0.05). Conclusion: Multi-b-value diffusion-weighted imaging scans can change dynamically evaluate human colon tumor xenograft model in nude mice after dru-g treatment, dynamic monitoring can be considered an early ant-iangiogenesis drug efficacy reference index.
