ApoEε4 Allele For Mild Cognitive Impairment And Alzheimer’s Disease Brain Microstructure Influence In Diffusion Kurtosis Imaging Research Study

Objective:Application of diffusion kurtosis tomography(DKI) magnetic resonance technology, comparison of Apo Eε4 allele carriers and noncarriers of cognitive function in normal elderlys, patients with mild cognitive impairment and Alzheimer’s disease brain differences in microstructure, explore the Apo Eε4 alleles on its brain microstructure change. Assessment of Apo Eε4 alleles and DKI technology in the application of cognitive function value for the early diagnosis of MCI and AD provide more valuable diagnostic basis.Methods:Selected 18 patients with AD(AD group),18 patients with MCI(MCI group)and cognitive function in normal elderly 18 cases(NC group), collection and its line of Apo Eε4 alleles of the detection of the oral mucosa, using chi-square test to three groups of Apo Eε4 alleles between carrying rate comparison uneffective routine MRI scans and DKI scanning at the same time, the measured bilateral frontal, parietal lobe,temporal lobe, corpus callosum, the value of the cingulate cortex and hippocampus after MK, MD value and FA value, using t test to each group in Apo Eε4 alleles are carriers and noncarriers DKI parameters measured by difference comparison.Results:1.AD group, the MCI group, and NC group differences in age, gender, education level has no statistical significance(P>0.05);2. AD group, MCI and NC group carrying rate of Apo Eε4 alleles between difference was statistically significant(P< 0.05), to carry the highest rate in the ADgroup and lowest in the normal group, MCI group is between in these two groups;3.AD group, the MCI group, and NC group each group of professionals to carry Apo Eε4 alleles of genome compared with not carry group.In the AD group, temporal lobe, the posterior cingulate, corpus callosum and hippocampus MK value decreases,and the difference is statistically significant(P< 0.05); In temporal lobe and hippocampus, the decrease of FA valuesand and the increase of MD values are statistically difference(P< 0.05);4.MCI group, carrying the Apo Eε4 genome compared with not carry group,temporal lobe, cingulate cortex and hippocampus of MK value decreases, and the difference is statistically significant(P< 0.05);Hippocampal FA value reduction, and statistically significant difference(P< 0.05);5. NC group, carrying the Apo Eε4 genome compared with not carry group,temporal lobe and hippocampus MK value decrease, and the difference is statistically significant(P< 0.05);Hippocampal FA value reduction, and the difference has statistical significance(P< 0.05)Conclusion:1. Apo Eε4 alleles in patients with AD carry rate is higher than normal elderly Apo Eε4 alleles of carrying rate;2. Carrying Apo Eε4 alleles of brain in patients with AD microstructure change more obvious,, DKI to early detection of such genes for microstructural changes of the brain, and provide more basis for early clinical diagnosis of the disease.
