Dividing The Trigeminal Nerve Of Trigeminal Neuralgia In Rats

Objective:1.Through the establishment of the trigeminal neuralgia model in rats.For analysis the trigeminal nerve,after the trigeminal nerve that have the rats of surgery treatment of the trigeminal neuralgia, it is about that the electrophysiological response and inflammatory factor at the distribution of nerve fiber.Evaluate about the effectiveness of the trigeminal nerve severed for postoperative pain relief.2.Study about the distribution characteristics of the third branch in trigeminal ganglia which go to reach the trigeminal nerve root.For clinical selective the trigeminal nerve severed operation,It is provides research basis and reduce the trigeminal nerve severed facial anesthesia and postoperative retinal ulcer.Method:1.The research choose 30 SD healthy male rats,With chronic narrow ring model to make the model of the trigeminal neuralgia,The subjects were divided into three groups randomly- infraorbital neuralgia, infraorbital nerve which was divided and the control group.At the two weeks,three weeks, four weeks,these three different group choose two rat from the group to get abdomen anesthesia and have hole craniotomy in the area of the contralateral VPM nuclear site.Let us use multi-channel electrophysiology monitor testing rats VPM nuclear to check changes in amplitude and frequency of discharge,and then open the chest,have the heart cavity perfusion,have fixed the brain tissue with 4%paraformaldehyde solution.Make the trigeminal ganglia tissue embed,section and use immunohistochemical method to test SP and CGRP which related to inflammatory factor expression and change.Through the immune microscope observation and statistical analysis.Judge the effect of the rats which trigeminal nerve was divided on the rats’ pain.2.Having injected fluorescence probe Dio, Dil, DId, into three nerve branches which is the infraobital nerve branch,the orbital nerve branch,the mandibular nerve branch and the spinal nucleus of the trigeminal nerve.Let the rats live in 72 hours and make the heart cavity infusion with 4% paraformaldehyde,have fixed the trigeminal nerve.Make the trigeminal ganglion nerve fiber for continuous frozen section,Use the fluorescence microscope to observe three branches characteristics and make image analysis.Result:1.Three experimental content contains SP and CGRP—the infraorbital neuralgia group,the group of infraorbital nerve which was divided whose contains SP was more higher than the control group.In four weeks later, the contents of SP was trend to reduce that is close to the control group.At the same time,the infraorbital neuralgia pain model is significantly higher than others.CGRP levels of them, the infraorbital neuralgia group,the group of infraorbital nerve which was divided,compared to the control group, is higher obviously.After four weeks away,that its CGRP showed that the trend of decline level is close to the control group.2.Electrophysiological experiment results is that the infraorbital neuralgia group have obviously higher than the the other groups significantly at discharge frequency and waveform amplitude.It is that the electrical signal of damage was recorded.Compared with the control group,at the early period,the group of the infraorbital nerve which was divided at discharge frequency and waveform amplitude is a bit higher.But the trend of wavefrom have no significantly difference with the control group,after the four week weeks.3.Through the fluorescent dyeing,It is found that the eye branch of trigeminal nerve lies in the inside area in trigeminal nerve.The area of infraorbital nerves is located in the eye of the lateral.Mandibular nerve distribution have located in the outside.In the trigeminal ganglia,the pselaphesia fibers and the nocicetive fibers start to separate and interlace with each others.The pselaphesia fibers basically focus on in the central trigeminalganglion,The located of the nocicetive fibers is decentralized comparactively which is focuses on the front of the line and go to the posterolateral.Conclusion:1.Compared the model of trigeminal neuralgia with the group of the trigeminal nerve which is divided,the control group,SP,CGRP, which related to the pain factor was decrease.Electrophysiological experiments show that trigeminal nerve which is divided at VPM nuclear site that Electrophysiological performance have similar with those of the control group,that have less pain electrical signal compared to the trigeminal neuralgia group.2.It is found that the eye branch of trigeminal nerve lies trigeminal nerve in the inside distribution area, the infraobital nerve is located in the middle,the mandibular nerve distribution in the outside area compared with the other branches in fluorescent dye.In trigeminal ganglia, three bundle fiber make the convergence of the different bundle depending on the type of fiber and reach the differential of the nucleus of trigeminal nerve which it rearrange the line at last.In the outside of the ganglion which lies the far side trigeminal ganglion,In that area,the spinal nucleus of fibers is relatively concentrated the outside of the fiber,which can choose as trigeminal nerve selective amputation site.
