Objective The rat model of trigeminal neuralgia was proformed by infraorbital nerve(ION)chronic constriction injury(CCI).Von Frey hairs test,Q-RT PCR,Western blot and immunohistochemical staining were used to investigate the effects on the expression of Pannexin-1(Panx 1)after injury of the trigeminal nerve in a rat model.Methods Adult male SD rats(n=45)were randomly divided into three groups:control group(Sham),CCI 1 week group and CCI 2 weeks group.After the trigeminal neuralgia rat model was constructed successfully,Von Frey hairs were used for the mechanical stimulation test on the ipsilateral whisker pad of rats.Q-RT PCR was used to detected the level of Panx 1 mRNA in the trigeminal ganglion(TG).Western blot was used to detected the expression of Panx 1 in the TG.The distribution of Panx 1 in the TG was detected by immunohistochemical staining.Results 1.Von Frey hairs test showed that the mechanical stimulation pain sensitivity threshold of rats in CCI group showed a significant decrease compared with Sham group.2.Compared with Sham group,the expression level of Panx 1 mRNA in TG of CCI group was significantly increased after ION-CCI.3.The expression level of Panx 1 protein in TG of CCI group was significantly enhanced after ION-CCI compared with Sham group.4.Immunohistochemical staining showed that after ION-CCI 2 weeks,the expression of Panx 1 protein in the trigeminal ganglia of CCI rats were significantly enhanced.Conclusions In the rat model of ION-CCI,the expression of Panx 1 in the TG was increased which indicated that Panx 1 may be participate in the occurrence and development of trigeminal neuralgia. |
