Objective To investigate the role of interleukin-17(IL-17) and interleukin-27(IL-27)in relapsing remitting multiple sclerosis(RRMS) and possible mechanismsin, and analysis of patients with multiple sclerosis cognitive impairment condition.Methods In a case-control study, plasma was collected from health examination center as healthy control group(n=45) and relapsing-remitting multiple sclerosis patients(n=45) from the Second Hospital of lanzhou University Department of Neurology from March 2010 to October 2015. The plasma levels of IL-17 and IL-27 were assessed by ELISA method, and combined with Expanded disability status scale(EDSS), Number of lesions of Gd-enhancing head(MRI), Mini-Mental State Examination(MMSE) and Montreal cognitive assessment scale(Mo CA) evaluation. Statistical analysis was performed with IBM SPSS 23,The comparison between the two groups using independent sample T test and comparing for paired samples T test within the group, and Before treatment variables by using Pearson correlation analysis.Results ⑴Plasma levels of IL-17 in before treatment were increased compared to the control group(P<0.05), Oppositely plasma IL-27 levels were significantly lower(P<0.05);Plasma concentrations of IL-17 in after treatment were decreased compared with before treatment,While a significant growth between plasma IL-27 concentration in after treatment and before treatment was found(P<0.05).⑵The EDSS was significantly improved in 4th weeks in patients’ group, but the number of lesions of Gd-enhancing was not significantly changed(P> 0.05).⑶The MMSE scores and the Mo CA scores were significantly decreased compared to the control group(P <0.05).⑷Plasma levels of IL-17 were negatively correlated to concentrations of IL-27 in patients with MS(r=-0.375); plasma levels of IL-17 were positively correlated to the EDSS score and the number of lesions of Gd-enhancing in MS((r= 0.360, r= 0.334); plasma levels of IL-27 were negatively correlated to the EDSS score, the number of lesions of Gd-enhancing, the MMSE and the Mo CA respectively(r=-0.319, r=-0.429, r=-0.356, r=-0.302); MMSE scores was positively correlated to the EDSS scores, the number of lesions of Gd-enhancing andDisease duration respectively( r=0.350, r=0.357, r=0.514); Mo CA scores was positively correlated to the EDSS scores, the number of lesions of Gd-enhancing and Disease duration respectively(r=0.362, r=0.321, r=0.538).Conclusion Our findings show IL-17 possesses proinflammatory properties, but IL-27 have antiinflammatory properties. As important of cytokines involve in the pathophysiological process of MS. We may regulate IL-17 and IL-27 can inhibit the inflammatory response in MS patients and improve symptoms and further found that Multiple sclerosis patients have cognitive impairment in the early.Therefore, They provide new ideas and direction for Multiple sclerosis pathogenesis,diagnosis and treatment.
