Objective:To investigate the prevention and treatment effects and preliminary discuss the action mechanisms of lycium barbarum polysaccharide(LBP) on hepatocyte steatosis model of hepatocyte in vitro.Methods:A model of non-alcoholic fatty liver disease(NAFLD) oleic acid-induced steatosis L-O2 strain hepatocytes(normal adult hepatocytes) was established. The model was devided into normal group steatosis model group prevention group and treatment group,The accumulation of lipid droplets in hepatocytes was observed under light microscopy after oil red-O staining after 48 hours. Detected the levels of alanine aminostransferase(ALT) aspartate aminostransferase(AST) triglyceride(TG) and the expression of m RNA and protein of adiponectin and resistin.Results:Much more obvious orange lipid droplets in hepatocytes were found in model group after cultured than in normal(P=0.000). Lipid droplets decreased in the prevention group and treatment group than in model group(P=0.000;P=0.000). Interfed in 10% LBP, the levels of ALT AST and TG were decreased(P=0.000;P=0.000;P=0.000); the expression of m RNA and protein of adiponectin were up-regulated(P=0.000;P=0.014) and the expression of m RNA and protein of resistin were down-regulated(P=0.000;P=0.000).Conclusion:Induced by expression of m RNA and protein of adiponectin and resistin,NAFLD can be prevented and treated effectivly by LBP.
