Objective: gingivitis model was established in rats and to observe the therapeutic effect of Radix Isatidis on the gingivitis animal model; Detect gene expression of rat gingival epithelial beta defense element 2 of SD, then study the relationship.of Radix Isatidis therapeutic effects and the induced expression of beta defensin Ang-2Methods: Forty healthy adult SD rats were selected(about 200g/only),then ligating rat maxillary first molar using orthodontic wire ligation method,to the side of the same teeth as control to establish rat gingivitis model; observed rats bilateral maxillary first molar gingival color,texture and shape by naked eye before the ligation,, measured and recorded the rats gingival index, sulcus depth. Successful modeling of 40 Wistar rats were randomly divided into positive control group(group A), negative control group(B group), middle dose group(C group that haploinsufficiency group)and high dose group(Group D, 2 times the dose group) each group had 10 rats. The positive control group(A group): 2mg/kg solution with LPS intravenous injection, the administration the next day; The negative control group(B group): normal saline 0.5ml/ intravenous injection, daily administration; The middle dose group(C group): to 2.1ml/kg traditional Chinese medicine monomer of Radix Isatidis injection, intravenous injection, daily administration; The high dose group(D group): to 4.2ml/kg traditional Chinese medicine monomer of Radix Isatidis injection, intravenous injection, daily administration; ligation wire was removed Before administration,color,texture and morphology of the rats maxillary first molar teeth were observed after 24 hours, measured and recorded the rat gingival index and sulcus depth. After continuous administration for 7 days and on the eighth day, rat gingival color, texture and shape were observed by naked eye, then measured and recorded the rat gingival index, sulcus depth; the rats were sacrificed and gingival tissue were removed,the gene expression level of rat beta defense element- 2 and protein was detected by real-time fluorescence quantitative PCR methodResults: 1, the observation of the rat gingival color, texture and shape, was detected in the rat gingival hemorrhage index and sulcus depth. The teeth the gingival inflammation successfully established a rat model of gingivitis; 2, to a week after the drug, rat gingival color, texture, and shape recovery, shallow depth of the gingival sulcus, gingival bleeding index was lower(p < 0.05); 3, high dose group, middle dose group, positive control group A beta- defense Ang-2 expression levels were significantly higher than those in negative control group(p < 0.05), but among the three groups compared no significant(p > 0.05).Conclusion: 1, Radix Isatidis on rat experimental gingivitis had a certain therapeutic effect; 2, Radix Isatidis on rat can induce the expression of gingival epithelial beta defensin Ang-2. 3, the therapeutic effect of Radix Isatidis may be related to its induction of beta defensin Ang-2.
