Objective:To measure the low density lipoprotein receptor related protein 6(LRP6)gene polymorphism,bone metabolism and bone mineral density(BMD)levels in postmenopausal women with type 2 diabetes mellitus(T2DM),further to explore their correlation.It lays the foundation for the identification and prevention of this population and the mechanism of this disease.Methods: 1.136 cases of postmenopausal women in Shihezi,Xinjiang,were divided into four groups: normal glucose tolerance(NGT)with normal bone mass(Agroup),T2 DM with normal bone mass(Bgroup),NGT with abnormal bone mass(C group),T2 DM with abnormal bone mass(D group),according to the results of medical history,OGTT and BMD.2.Determination of biochemical indexes: Hemoglobin A lc(Hb A1c)was measured with high performance liquid chromatography,alkaline phosphatase(ALP),Ca,P,triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),fasting plasma glucose(FPG)measured with biochemical analyzer measured directly.3.The BMD levels of the L1-4 and the femoral neck were detected by dual energy X-ray(DEXA)bone densitometer.4.DNA was extracted from the peripheral venous blood samples from the participants.The genetic polymorphism of LRP6 gene rs10743980,rs2417086,rs1181334 and rs2302685 loci was detected by time of flight mass spectrometry(TOF MS).The detection of SNP was carried out and the distribution of genotypes between groups was analyzed and compared.5.The genotype distribution was tested by the chi-square test of genetic balance,to determine whether it was consistent with the Hardy-Weinberg balance.6.The SPSS19.0 statistical software was used for data processing.The variables were expressed in the form of sx ±.If the baseline data were all tested by t test,the difference between two groups was analyzed by variance analysis,P < 0.05 revels that the difference was statistically significant.Results:1.The distribution of rs10743980,rs2417086,rs1181334 and rs2302685 gene polymorphisms of LRP6 gene in postmenopausal women in Shihezi District of Xinjiang is not statistically different in groups with different bone mass and glucose metabolism.2.Compared with A group,FPG level are higher in B and Dgroups(5.18±2.13,7.60±2.13,7.50±2.13),Hb A1 c levels are higher in B and Dgroups(5.92±1.06,7.19±1.05,7.50±1.05),(P< 0.05);The BMD levels of L1-4,femoral neck in A groups are higher than those in C and D groups,L1-4(1.19±0.15,0.85±0.14,0.91±0.15),femoral neck(0.88±0.18,0.72±0.19,0.75±0.19),TG levels are lower in C and D groups(2.50±1.52,1.51±1.50,1.70±1.52),P <0.05.There was no significant difference among the other groups and metabolic indexes.3.The genotype distribution of rs10743980,rs2417086,rs1181334 and rs2302685 of LRP6 gene conforms to the Hardy-Weinberg genetic balance,(P >0.05).4.There was no statistical difference in the distribution of LRP6 gene genotype between the four groups.5.Each of LRP6 locus of two different genotypes of wild type(homozygous)and mutant(heterozygous /homozygous)compared with clinical data and metabolic indexes:comparing the polymorphism of rs10743980 withthe postmenopausal women of T2 DM with abnormal bone mass,there was statistically significant differences in HDL-C(P=0.03)and Hb A1c(P=0.04).The level of Hb A1 c in mutant group(CT/TT)is higher than it in wild type(CC),the level of HDL-C in mutation type(CT/TT)was lower than it in wild type(CC);Analyzing the polymorphism of rs2417086 and T2DM in postmenopausal women with abnormal bone mass,there was statistical difference in HDL-C(P=0.03)and Hb A1c(P=0.04).The level of Hb A1 c in mutant group(AG/GG)is higher than it in wild type(AA),the level of HDL-C in mutation type(AG/GG)was lower than it in wild type(AA).Conclusion: 1.The distribution of rs10743980,rs2417086,rs1181334 and rs2302685 gene polymorphisms of LRP6 gene in 136 cases of postmenopausal women in Shihezi District in Xinjiang had no difference with bone mass metabolism.2.The mutation of LRP6 gene rs10743980 and rs2417086 loci may be related to thebone mass in patients,T2 DMin postmenopausal women in Shihezi,with abnormal lipid metabolism and glucose metabolism disorders.
