The Experimental Study On The Relationship Between The Effects Of Drug Compatibility And Pharmacodynamic Of The Herbal Pairs Of Ge- Ntiana Macrophylla Pall In Treating RA Rat Models Of Wind-cold-dampness Arthralgia

Objective Gentiana macrophylla was used as basic medicine,cooperating with Clematis chinensis Osbeck, Taxillus chinensis(DC.) Danser and Stephania tetrandra to form the compatibility of mild-and-warm(Gentianae macrophyllae-C lematis chinensis Osbeck, the compatibility of mild-and-mild(Gentianae macrophyllae- Taxillus chinensis(DC.) Danser), and the compatibility of mild-and-cold(Gentianae macrophyllae-Stephania tetrandra). Through studying different effects of different compatibility of Gentianae macriphyllae on rheumatoid arthritis model rats of wind-cold-dampness arthralgia, the relationship between peoperties and efficacy, properties and effect of Chinese medicinal herbs were explored.Methods:Rheumatoid arthritis rat models of wind-cold-dampness arthralgia which had the Traditional C hinese Medicine Arthralgia of combining disease and syndrome was built, by the model rats general state, joint histomorphology and quantitative evaluation of arthritis,combination with the index of immune organs and detection the level of TNF-α, IL-1, CCP, Ig G, PGE2 and LDH in serum, synthetically analyze the relationship between compatibility of different properties of Chinese medicinal herbs and effect of different compatibility in the syndrom of wind-cold-dampness arthralgia.Results: 1) Observation the foot metatarsus swelling degree of rats, compared with t he model group of combining disease and syndrom, the treatment group had relieved the foot metatarsus swelling degree, the effect of the Gentianae macrophyllae- Taxillus chinensis(DC.) Danser group and the Gentianae macrophyllae- Clematis chinensis Osbeck group was obvious. 2) the study of rats immune organ index showed that the thymus index of every group hadn’t statistical difference; the spleen index of Gentianae macrophyllae-Stephania tetrandra group and single Gentiana macrophylla group significantly increased(P<0.01).3) The results of rats ankle tissue morphology showed that single Gentiana macrophylla group and the Gentianae macrophyllae- Clematis chinensis Osbeck group significantly reduced the synovial proliferation and angiogenesis. 4) The factor of rats serum related research results show that :1 Compared with the blank group, the TNF-α and IL-1β of wind-cold-dampness model group of combining disease and syndrom creased significantly at the same time(P< 0.01);Compared with model group, the TNF-α of wind-cold-dampness model group of combining disease and syndrom significantly increased(P<0.01), suggesting the climate factors of wind-cold-dampness can increase the RA condition; Compared with the wind-cold-dampness model group of disease syndrome, the level of TNF-α and IL-1β reduced in treatment groups, showed that Tripterygium wilfordii,single Gentiana macrophylla and the different compatible of Gentiana macrophylla has a therapeutic effect on RA model rats. 2 Compared with the wind-cold-dampness model group of combining disease and syndrome, the CCP of every Chinese Traditional Medicine group were reduced, there exists a significant difference in the Gentianae macrophyllae- Clematis chinensis Osbeck group(the compatibility of mild-and-warm)(P<0.01), but also reflect the effects of cold syndrome with hot medicine.Compared with the wind-cold-dampness model group of combing disease and syndrome, the Gentianae macrophyllae- Taxillus chinensis(DC.) Danser group(the compatibility of mild-and- mild), the Gentianae macrophyllae-Stephania tetrandra group(the compatibility of mild- and-cold) had the higher levels of Ig G, there exists a significant difference(P<0.05).In cold syndrome cases, the compatibility of mild-and- mild and mild-and-cold didn’t relieve syndrome or even worse conditio n.3 Compared with the wind-cold-dampness model group of combining disease and syndrome, the Gentianae macrophyllae-Clematis chinensis Osbeck group(the compatibility of mild-and-warm),the Gentianae macrophyllae- Taxillus chinensis(DC.) Danser group(the compatibility of mild-and- mild) reduced the level of LDH, the difference is significant(P<0.01), The LDH level of the positive group and the single Gentiana macrophylla also reduced, the significant difference existed(P<0.05). the compatibility of mild-and-warm can improve the illness of the syndrom of Wind-Cold-Dampness arthralgia.Conclusion: In the RA model rats of wind-cold-dampness arthralgia, compared with model group of combing disease and syndrome, the Gentianae macrophyllae-Clematis chinensis Osbeck group(the compatibility of mild-and-warm) had good effect, the Gentianae macrophyllae- Taxillus chinensis(DC.) Danser group(the compatibility of mild-and- mild)(suitable) peace times, single Gentiana macrophylla group, Gentianae macrophyllae-Stephania tetrandra group(the compatibility of mild-and-cold) were worser."Heat syndrome with cold property medicine, cold syndrome with warm property medicine,this treatment principle is scientific and effective.
