Purpose:To investigate the HIV-derived novel miRNA99 whether can induce macrophages to release TNF-α,IL-6,IL-1β and regulate the phagocytosis of macrophages.Methods:The level of TNF-α,IL-6,IL-1β released by macrophages challenged with miRNAs(mi RNA99 、 miRNA-TAR) 、 Lipid A were measured by ELISA.The phagocytosis of macrophages was measured by Confocal microscopy and flow cytometry.Results:THP-1 challenged with PMA(100ng / ml)for 24 hours became smooth adherent,and the expression of CD11 b of macrophages was up to 99%. Macrophages release elevated levels of TNF-α, IL-6, IL-1β in Lipid A group and miRNA99 group, but not in miRNA-TAR group,and the level of miRNA99 group was much higher than the level of LipidA group. The phagocytic capabilities of the three groups of macrophages were declined,especial in the group of miRNA99.Conclusion :1.miRNA99 stimulated macrophages released increased levels of TNF-α, IL-6,IL-1β;2.miRNA99 made the phagocytosis capabilities of macrophages decline.
