Objective:To explore the effects of thymopentin(TP-5) on the phagocytosis of rat enterocoelia macrophages in vitro.To study whether the TP-5 act on the phagocytosis of rat enterocoelia macrophages by influence the Staphylococcus aureus.Method:(1)To obtain macrophages:rats were injected with 5% starch solution 72 hours ago for generating and activating macrophages.Then lavage and fetch enterocoelia macrophages.(2)Part one:The Staphylococcus aureus mixed with TP-5 30 minutes, then the mixture added in the macrophage solution.After 40 minutes,the liquid was smeared and fixed on the glass slides.Part two:①Concentration gradient experiment:different concentration of TP-5 (0mg/ml、0.2mg/ml、1mg/ml、5mg/ml)mixed with the Staphylococcus aureus.After 30 minutes,the mixture were added in the peritoneal lavage fluids for 40 minutes.And the mixture solution was smeared and fixed on the glass slides,which were stained with Giemsa staining, to observe the results with the microscope.②Time gradient experiment:1mg/ml of TP-5 mixed with the Staphylococcus aureus. After 30 minutes, the liquid were added in the peritoneal lavage fluids,and the mixture solution was smeared and fixed on the glass slides in 5 minutes、10 minutes、20 minutes、40 minutes、80 minutes,which were stained with Giemsa staining to observe the results with the microscope.(3)Phagocytosis rate and phagocytosis number were reflected the effect of thymopentin on the phagocytosis of rat enterocoelia macrophages in vitro.Results:(1)Part one,the phagocytosis rate of control groups was 40.13±6.71%,while the experimental groups was 46.90±4.49%.Meanwhile, the average of phagocytosis number of control groups was 3.59±0.73,and the experimental groups was 4.51±0.90.Comparing with the control groups,the average of phagocytosis rate and phagocytosis number of experimental groups both improved significantly (p< 0.01).(2)Part two,after mixed with different concentration of thymopentin (TP-5) and acted on different times,the cellular morphology and the phagocytosis rate were different.①Concentration gradient experiment:the phagocytosis rate and phagocytosis number of lmg/ml group was higher them other groups (0mg/ml、0.2mg/ml、5mg/ml) during the same time, while the phagocytosis rate and phagocytosis number of the 5mg/ml group were the lowest.②Time gradient experiment:After 5 minutes、10 minutes、20 minutes、40 minutes、80 minutes,the phagocytosis rate and phagocytosis number were presented with progressive tendency. And the macrophages ruptured and the number reduced in 80 minutes group.Conclusion:①T-5 could improve the phagocytosis of rat enterocoelia macrophages by acting on Staphylococcus aureus.②TP-5 acting on the influence of macrophage phagocytosis is related to concentration.In certain range of concentration, the increasing of the concentration of the TP-5,macrophage phagocytosis rate and phagocytosis number are increased. ③The function of macrophage phagocytosis is related to the time.With the passage of time, the phagocytosis rate and phagocytosis number are increased,and showed an increasing tendency.
