Objective:1. Taking the autophagy marker protein(Microtubule-associate Protein LC3-Ⅱ,Beclin-1) as the observation target and applying the method of immunohistochemical staining, this paper studies the corresponding expression and significance during the process of autophagy in cervical intraepithelial neoplasia and cervical squamous cell carcinoma tissues pertaining to different pathological grades.2. After the case analysis of clinical data regarding to HPV infection status and examining its influence on the process of autophagy, this paper discusses the changing effect of autophagy in cervical intraepithelial neoplasia and cervical squamous cell carcinoma tissues.Methods:1. Select Jiexiu area of cervical cancer screening crowd scene and the second hospital of Shanxi Medical University, from June 2014 to June 2015,18-65 year old female line of cervical cancer screening officers to randomly from chronic inflammatory disease detection, CIN1, CIN2, CIN3, cervical squamous cell carcinoma extract all 30 cases included in the study for a total of 150 people.2. Select paraffin sections in tissues clearly diagnosed as cervical chronic inflammation, CIN1,CIN2/3 and cervical squamous cell cancer, conduct immunohistochemical staining of LC3-Ⅱ and Becin-1 on 30 slices from each tissue and divide them into groups according to the semi-quantitative scores obtained through examination under the microscope.3.Using SPSS 13.0 for statistical analysis, comparison between data sets serving grade chi-square test; usage trends trends between groups chi-square test, correlation between groups using Kappa test, test level α <0.05.Rseults:1. The autophagy protein of LC3-Ⅱ shows different positive rates: 83.3%(25/30) in normal cervical tissue, 73.3%(22/30) in CIN1, 63.3%(38/60) in CIN2/3 and46.7%(14/30) in squamous cell carcinoma. In the same manner, Beclin-1 sees a similar trend: 90.0%(27/30)in normal cervical tissue, 83.3%(25/30) in the CIN1 group, 60.0%(36/60) in the CIN2/3 group, and 53.3%(16/30) in squamous cell carcinoma. In a word, the positive expression rate declines as the lesion grade ascends, while the negative expression shows an opposite trend.2. HR-HPV infection may weaken the positive expression of LC3-Ⅱ and Beclin-1(P<0.05), excepting the CIN2/3 group based on the index of LC3-Ⅱ and the CIN1 group based on Beclin-1.3. LC3-Ⅱ and Beclin-1enjoys a similar expression model in tissues of intraepithelial lesion and cervical squamous cell cancer.Conclusion:1. As observation targets of autophagy strength, LC3-Ⅱ and Beclin-1 both prove the same declining trend along with the deterioration process from cervical intraepithelial neoplasia to cervical cancer.2. The mechanism of autophagy in the uninfected group works better than the HPV infected group.3. LC3-Ⅱ and Beclin-1enjoys a similar expression model in tissues of intraepithelial lesion and cervical cell cancer. |
