Objective:To investigate the effects and possible mechanisms of Xuezhiqing prevention of atherosclerosis model in rats, and to provide scientific, objective basis for anti-AS of Xuezhiqing.Methods:Rats were randomly divided into four groups, including normal control group (group A, n=6) to the normal diet for 16 weeks, and model group (group B, n=10), rosuvastatin group (group C, n=10), and Xuezhiqing group (group D, n=10) to the High fat diet for 12 weeks. On the first day of feeding, group B, group C, and groupD were disposably injected vitamin D3 60 Wan U·1kg-1 into intraperitoneal to use aseptic technique.In the fourth weekend after the feeding, group B, group C, and groupD were disposably injected vitamin D3 10 Wan U·1kg-1 into intraperitoneal to use aseptic technique once more. In the 12th weekend, two rats randomly selected from group B, C and D were killed, and artery was detected pathology staining under microscope, which displayed AS plaque formation, confirming a successful atherosclerosis model in rat.Then we entered the treatment phase; group C with rosuvastatin 1.05mg/kg·d one time everyday, group D were gavaged with Xuezhiqing 8.19g/kg·d one time everyday,, groupA and groupB were gavaged with the same volume of distilled water one time everyday, all groups were gavaged four weeks. After 4 weeks, serum lipids in rats of all four groups taken from Inferior vena cava under anesthesia,were measured with the concentrations of TG、TC、 LDL-C、HDL-C、IL-6 and TNF-α,and then taking part of the thoracic aorta to detect pathological staining and MMP9 immunohistochemical staining.Results:1.Compared with group A, TG in group B was higher, having Statistical significance(P <0.05), TC in group B was significantly higher, HDL in group B was significantly lower, and LDL in group B was significantly higher, having Statistical significance(P<0.01); Compared with group B, TG in group C and group D were lower, having Statistical significance(P<0.05), TG was significantly lower, HDL-C was significantly higher, and LDL-C was significantly lower, having Statistical significance(P<0.01).2.Compared with group A, IL-6 and TNF-a in group B were significantly higher, having Statistical significance(P<0.01); Compared with group B, IL-6 and TNF-a in group C were significantly lower, having Statistical significance(P<0.01); Compared with group B, IL-6 in group D were significantly lower, having Statistical significance(P<0.01), and TNF-a was significantly lower, having Statistical significance(P<0.05).; Compared with group D, TNF-α in group C was significantly lower, having Statistical significance(P<0.01).3.Compared with groupA. thoracic aortic pathology staining in group B were displaying obvious visible intima lipid plaque formation, vascular endothelial cell damage, foam cell formation, smooth muscle cell migration and proliferation; compared with group B, thoracic aortic pathology staining in group C and group D displyed smooth endometrial, vascular endothelial cells beginning to repair, migration and proliferation of smooth muscle cell being not obvious and the more lipid plaque regressing obviously.4.MMP-9 expression in group A was not obvious; MMP-9 expression in group B was strongly positive; MMP-9 expression in group C and D was a weakly positive expression.Conclusion:The anti-AS effect of Xuezhiqing is concerned about regulating the blood lipids, anti-inflammatory effects and reducing the expression of MMP-9 in artery.
