ObjectiveEpicardial adipose tissue(EAT) mainly distributes in atrioventricular groove, interventricular groove, right ventricular free wall and left ventricular apex.There is no fascia tissue between EAT and the wall of coronary artery.High density adipose tissue contracts with coronary artery system directly.For men and women after menopause,the maintenance of serum estrogen level mainly depends on the transformational ability of aromatase which can convert androgen into estrogen to effect local or adjecent tissues and organs,then play a role to protect coronary artery.However the reaserch about the protective effects of estrogen resulting from EAT and which has more important effect comparing estrogen in plasma and EAT,at present,there have no ralevent research.Accroding to measuring and comparing the the expression level of aromatase mRNA in EAT between CHD and non-CHD patients,we aimed to try to explain the functions and influences of aromatase in EAT to CHD.MethodsBefore cardiopulmonary bypass,5-10ml venous blood was taken from patients, get serum by using centrifugal machine,then put the serum into refrigerator setting in -20℃. The serum estrogen levels is measured by chemiluminescence determination.The EAT (5-10mg) get from right atrioventricular groove, initiation part of right coronary artery before cardiopulmonary bypass.The expression of aromatase mRNA is determined by rt-PCR.And the quantity of the aromatase protein is determined by ELISA.The difference of the level of expression of aromatase mRNA and quantity of aromatase protein between CHD and non-CHD patients is gotten by statistical methods.ResultsThe serum estrogen levels have no difference between CHD and non-CHD patients.The level of the expression of aromatase mRNA and the quantity of aromatase protein in CHD patients’ EAT is lower than non-CHD patients.Conclusions1.The aromatase can be tested in the EAT.The EAT has the transformable function of sex hormone.2.The vessel injury in atherosclerosis of coronary artery may derive from the decrease of the expression of aromatase in EAT.
