Association Between MicroRNA-208a、210、506 And Acute Myocardial Infarction

Objective:The expression levels of miR-208a, miR-210 and miR-506 in the plasma of acute myocardial infarction patients and healthy control population were detected by the method of real-time fluorescent quantitative polymerase chain reaction. It explored the assosiation between miR-208a, miR-210, miR-506 and the severity of coronary atherosclerosis, compared the value of their diagnosis of acute myocardial infarction, and further explored the expression differences between the Korean-Chinese nationality and Han nationality of patients with AMI in Yanbian area.Methods:This study included a total of 77 subjects who visited The Affiliated Hospital of Yanbian University from April 2014 to September 2014 because of chest pain and hospitalized in the Department of cardiology.The experimental group was divided into acute myocardial infarction (AMI) group (50 cases) and control group (27 cases).AMI group was divided into emergency surgery subgroup (15 cases) and selective operation subgroup (35 cases), Korean-Chinese nationality (20 cases) and Han nationality (30 cases). In addition, according to the Gensini scores, the integral was 0 (27 cases), integral< 30 (13 cases), the integral is equal to or lower than 30 to less than or equal to 60 (24 cases), integral> 60 (13 cases). The expression levels of miRNA-208a, miRNA-210 and miRNA-506 in plasma were detected by the method of RT-PCR. The contents of cardiac troponin I (cTnl) and creatine kinase isoenzyme (CK-MB) were detected by the automatic biochemical analyzer. Finally, the detection results and clinical data were analyzed statistically.Results:The relative expression levels of miR-208a and miR-506 in AMI group was higher than that in control group (P<0.01, P<0.05), and the relative expression level of miR-210 was lower than that of the control group (P<0.01). The relative expression level of miR-208a in emergency operation group was higher than the elective operation group (P<0.05).Three miRNAs’relative expression levels between the Korean-Chinese nationality and Han nationality were without significant difference (P>0.05), and did not differ between the Korean-Chinese nationality and Han nationality in the control group and the AMI group(P>0.05). The relative expression of miR-208a was significantly positively correlated with age, CK-MB, cTnl and pro-BNP (r=0.254 P<0.05, r=0388 P<0.01, r=0.495 P<0.01, r=0.405 P<0.01). The relative expression of miR-210 was negatively correlated with age and pro-BNP (r=-0.252 P<0.05, r=-0.292 P<0.05). The relative expression of miR-506 was positively correlated with LDL-C, CK-MB, cTnl and pro-BNP (r=0.230 P<0.05 r=0.277 P<0.05, r=0.331 P<0.01, r=0.249 P<0.01). Gensini score was significantly positively correlated with miR-208a and miR-506(r=0.682 P<0.01, r=0.353 P<0.01), while was significantly negatively correlated with miR-210 (r=-0.416 P<0.01). The areas under the ROC curve of miR-208a, miR-210, miR-506, cTnl and CK-MB were 0.917(95%CI 0.856-0.979),0.753(95% CI 0.624-0.882),0.807(95% CI 0.699-0.915), 0.758(95%CI 0.653-0.864),0.750(95%CI0.644-0.856).Conclusion:1. MiR-208a could be used as a biomarker for early diagnosis of AMI, but its diagnostic efficacy is not better than CK-MB, cTnl; miR-210, miR-506 may also be a diagnostic marker of AMI, but their early diagnostic abilities may be less than miR-208a;2. MiR-208a has the potential to respond to the severity and risk stratification of coronary artery atherosclerosis;3. The levels of miR-208a, miR-210, miR-506’s expressions in plasma of AMI patients don’t have regional and ethnic differences.
