| AIM:To explore the Effects of Herba Cistanches on lung immune function in rats with tracheostomy tube through the detections of Pulmonary beta-defensins-2(BD-2),Surfactant Associated Protein A (SP-A),secretory immunoglo bulin A (SIgA),and serum Interleukin 2(IL-2).METHODS:All of 54 male SD rats were randomly divided into 3 groups:normal control(A),model with tracheostomy tube(B),and the Herba Cistanches-treatment group (C),group C were treated with Herba Cistanches decoction and group A and B were treated with physiological saline after 6 hours from models built.Every 6 rats of each group were killed at 24h,72h,168h,and 2ml blood taken from aortaventralis was used for detecting the concentration of IL-2 by IL-2 ELISA Kit, Ligation of right main bronchus and took 100mg right lung tissues to detect the expression of BD-2 by RT-PCR technology, the rest of right lung tissues were sent to take Pathological examination immediately, Smith scores was used to evaluate the pulmonary injuries.The concentration of SP-A and SIgA in BALF taken from the left lung were detected with ELISA method by SP-A ELISA Kit and SIgA ELISA Kit.RESULTS:(1) The observation of lung tissue pathology were found normal in group A,and injuries such as edema, alveolar and interstitial inflammation, alveolar and interstitial hemorrhage, atelectasis were observed in group B and C,the Smith scores in group B and group C were higher than group A,the injuries of lung tissues examined at three phases(24h,72h,168h) were different from every other phases in the group B and C,they became severer as the time went on.There were no significant differences between smith’s scores in group C and smith’s scores in group B in the counterpart phase. (2) The expressions of BD-2 in lung tissues increased obviously in 24h after the rats were modeled with tracheostomy tube, then decreased gradually in 72h, and reached the lowest level in 168h, when treated with Herba Cistanches, the expressions started increasing obviously in 24h, then turned to decrease in 72h, and got to lowest in 168h, but their levels still higher than group A and group B. (3) The concentrations of SP-A in BALF in group B and C were Significantly higher than group A at 168h,and highest at 24h,lowest at 168h,they became more and more lower as the time went on.The level in group C were higher than group A at 24h,72h and 168h. (4) The concentrations of SIgA in BALF in group B were significantly higher than group A at 24h,72h and 168h,and highest at 72h,then gradually went down,and reached lowest at 168h,while treated with Herba Cistanches,the concentrations of SIgA were more higher than group A and group B at 72h and 168h. (5) The levels of serum IL-2 didn’t change much after operate with tracheostomy tube,but they increased significantly in 72h and 168h after being treated by Herba Cistanches.CONCLUSIONrtracheostomy tube on rat may lead to lung injuries,such as edema, alveolar and interstitial inflammation, alveolar and interstitial hemorrhage, atelectasis,and also lead to immunologic derangement,maybe,because of self-protective reaction,the expressions of BD-2 and the concentrations of SP-A,SIgA increased in the early phase vicariously,then turned to decrease gradually,And Herba Cistanches shows its protective function to the lung through improving the level of BD-2,SP-A,SIgA,and enhance to body immune through improving the serum IL-2. |
