Cistanche Herba is derived from the dry fleshy stem of C.deserticola which belongs to orobanchaceae plants.Because it is produced in the desert of China,C.deserticola has very high medicinal value and is one of the precious traditional Chinese medicinal materials in the sand region,it is known as the Desert Ginseng.It is Mainly for the treatment of yang deficiency,constipation and so on.Cynomorium is one of the rare and precious Chinese herbal medicines in the desert region as similar efficacy as C.deserticola,but the market price gap between the two is relatively large.The price of C.deserticola is about ten times that of Cynomorium,but the comparative study is still very little about efficacy of them.In this paper,C.deserticola as the main research object,by high performance liquid chromatography to test C.deserticola components of different years,using water extract of C.deserticola of different years takes gavage test on mice in order to explore differences in efficacy on male and female small rats under the different growth period and different doses.By the combination of intragastric administration of Cistanches deserticola Y.C.Ma and Cynomorium songaricum Rupr and the combination of the two to compare the efficacy of C.deserticola and Cynomorium on male and female mice.During the gavage process,low,medium,and high doses were set for each medicinal material,and low doses and high doses are not within the range of medicinal herbs prescribed by the Chinese Pharmacopoeia.The main research conclusions are summarized as follows:1.There are regular differences in the content of components in different years of growth C.deserticolaThe content of echinacoside in C.deserticola from one year to four years is respectively34.33±0.114mg/g?9.04±0.019mg/g?10.42±0.037mg/g?37.12±0.152mg/g,indicating that the content of echinacoside in four years old C.deserticola is the highest,followed by one-year,while the content of the two-year raw C.deserticola is the lowest.The content of verbascoside in C.deserticola from one year to four years is respectively 0.90±0.016mg/g ?0.45±0.002mg/g ? 5.12±0.091mg/g ? 1.71±0.022mg/g,indicating that the content of verbascoside in three years is the highest,followed by four years,while the content of verbascoside in two years is the lowest.The content of tannin in C.deserticola from one year to four years is respectively 18.56±0.136mg/g,11.27±0.102mg/g,4.26±0.069mg/g,and8.93±0.178mg/g,indicating that the content of tannin in one year is the highest,followed by two years and the lowest content is in three years.The content of catechinic acid in C.deserticola from one year to four years is respectively 17.79±0.156 ?g/g,18.30±0.188 ?g/g,18.68±0.175 ?g/g,and 16.11±0.134 ?g/g,indicating that the content of three years is the highest,followed by two years and the content is the lowest in four years.The highest annual content of echinacoside and tannin in C.deserticola shows that the growth cycle of C.deserticola is three years.2.Significant differences in the efficacy of male and female mice between the years of growth and the doses of C.deserticolaFor female mice,the four-year low-dose group has the most significant memory improvement,the four-year middle-dose group has the worst effect on memory improvement,the four-year low-dose group of C.deserticola has the most significant anti-fatigue effects,the low-dose group had the worst anti-fatigue effect,the two-year high-dose group has the strongest anti-oxidation effect,the three-year low-dose group has the weakest anti-oxidation effect,the one-year-old low dose of C.deserticola is the lowes in weight gain.The three-year-old medium-dose C.deserticola medium dose group and the four-year medium-dose C.deserticola medium-dose group has the highest weight gain values.In different doses,but in the same years,the weight gain values ranged from low to high,followed by low-dose group,high-dose group and middle dose group,annual low dose of C.deserticola group has the strongest laxative effect on mice,and two-year middle dose group has the weakest laxative effect on mice.For male mice,the annual low-dose group has the most significant improvement in memory.The three-year middle-dose group has the worst effect on memory improvement,the one-year low-dose group has the best anti-fatigue effect,and the three-year middle-dose group has the same effect.The anti-fatigue effect is the weakest;the four-year low-dose group has the strongest anti-oxidation effect,and the two-year middle-dose group has the weakest antioxidant effect;the three-year-old low-dose group of C.deserticola gives the lowest weight gain value to mice.In the four-year-old high-dose C.deserticola group,the weight gain value of the mice is the highest;the low-dose C.deserticola three-year group has the strongest laxative effect on the mice,and the high-dose C.deserticola four-year-old group has the weakest laxative effect on the mice.3.Within the range of dosages stipulated in the Chinese Pharmacopoeia,the efficacy of C.deserticola in different ages for male and female mice is different.For female mice,one year old C.deserticola has the best effect on improving the memory.The four-year-old C.deserticola has the worst effect on mice's memory improvement.The three-year-old C.deserticola has the strongest anti-fatigue effect.One year old C.deserticolahas anti-fatigue effect on mice.The effect is the weakest;the three-year-old C.deserticola has the best anti-aging effects on mice,and the two-year-old C.deserticola has the worst anti-aging effect on mice;the one-year-old C.deserticola has the smallest body weight gain,and the two-year-old C.deserticola has the largest increase in body weight.One year old C.deserticola has the strongest defecation effect on mice,and two-year old C.deserticola has the weakest laxative effect on mice.For male mice,the four-year-old C.deserticola has the strongest effect on memory improvement.Three-year-old C.deserticola has the worst effect on mice's memory improvement;two-year-old C.deserticola has the best anti-fatigue effect on mice,The anti-fatigue effect of three-year-old C.deserticola is the worst;the three-year-old C.deserticola has the best anti-aging effect,and the two-year-old C.deserticola has the worst anti-aging effect on mice;the three-year-old C.deserticola has the smallest increase in body weight,for two-year-old C.deserticola and four-year-old C.deserticola,the weight gain value of the male mice is the largest;the three-year-old C.deserticola has the strongest effect on the defecation of mice,and the four-year-old C.deserticola has the weakest laxative effect on the mice.Through the comparison of male and female,it is found that C.deserticola takes better efficacy to females in improving memory,anti-fatigue and anti-aging than males.In body weight and laxative tests,it is found that the weight gain values of mice are related to the intensity of laxative effects.When the laxative effect is stronger,the weight gain value of mice is smaller,and when the laxative effect is weaker,the weight gain of rats is greater.4.The drug efficacy of C.deserticola and Cynomorium under mixture of them or using them respectively and the different gavage doses on male and female mice is different.In the female group,the memory test finds that the middle dose of Cynomorium has the most significant effect on the memory improvement of mice,and the high dose of Cynomorium has the weakest effect on the improvement of mice's memory;In the anti-fatigue experiment,the high dose of C.deserticola to mice is the most significant in the improvement of endurance,and the lowest dose of Cynomorium is the worst.In the antioxidant test,the mice in the low-dose C.deserticola group has the most significant effect,and the low-dose Cynomorium has the worst effect.In the weight and laxative tests,it is found that the effect of the high dose of the mixed solution is the most significant,and the efficacy of the medium dose of C.deserticola is the worst.In the male group,the memory test finds that the mixed solution dose has the most significant effect on the improvement of memory in mice.The high dose of Cynomorium hasthe weakest effect on the improvement of mice's memory.In the anti-fatigue test,in the low dose of C.deserticola on mice,the most significant increase in endurance is achieved,and the lowest dose of Cynomorium is the worst.In the antioxidant test,the mice in the medium dose group of C.deserticola shows the most significant effect,and the Cynomorium high dose has the worst effect;they are found in body weight and laxative tests.High-dose Cynomorium has the most significant effect,and C.deserticola's mid-dose has the worst efficacy.5.Within the range of dosages specified in the “Chinese Pharmacopoeia”,the effects produced by mixture of C.deserticola and Cynomorium or using respectively are different on male and female mice.In the female group,in the memory test,the effect from strong to weak is successively Cynomorium group,mixed liquor group and C.deserticola group.In the anti-fatigue experiment,the effect from strong to weak is successively the mixture group,C.deserticola group and Cynomorium group.Among the antioxidant assays,the effect ranged from strong to weak is successively C.deserticola group,mixed liquor group and C.deserticola group.In mice body weight and laxative tests,the efficacy from strong to weak is found to be Cynomorium group,mixed liquor group and C.deserticola group in proper order.In the male group,the memory test finds that the effect from strong to weak is successively the mixture group,C.deserticola group and Cynomorium group.In the anti-fatigue test,the effect is consistent with the female group.In the antioxidant assay,the effect is the strength which is consistent with that of the female group.In the weight and laxative tests of the mice,the effect is the same as that of the female.Through the comparison of male and female mice,it is found both the mixture of C.deserticola and Cynomorium and using them alone have stronger effects on females in improving memory,anti-fatigue and anti-aging than males.In the memory test,it is found that the efficacy of the medium doses for female mice is stronger than that of low doses and high doses,but for the male,the effect of low doses is stronger than medium doses and high doses.In the anti-fatigue test and body weight and laxative tests,it is found that the efficacy of the mixed liquid is stronger than that used alone,indicating that C.deserticola and Cynomorium have a synergistic effect on fatigue resistance,body weight and laxativeness for little mice.
