Objective:This study used Taishan Panshi San which has the function of tonifying qi and invigorating spleen and Yangxue Antai to intervene in SD rats model of abortion, to observe the changes of cytokine IFN-y, IL-2, IL-10 in pregnant rat’s serum and expression of TNF-a in deciduas’tissues of maternal-fetal interface, so it can provide the experimental basis for the mechanism of Taishan Panshi San in the treatment of abortion, which lays the theoretical foundation for the development of Chinese medicine for the treatment of abortion.Methods:15 SPF grade male SD rats and 30 SPF grade female SD rats were selected, and then put 1 male SD rat and 2 female SD rats into a cage. The pregnant rats were randomly divided into 3 groups:normal group, model group and intervention group, each group of 10 pregnant rats. The normal group of pregnant rats was given distilled water to 10ml/kg/d during 1 to 11 days of gestation, lml/kg/d of ethanol was injected subcutaneously during 6 to 8 days of gestation. Pregnant rats of the model group was given distilled water 10ml/kg/d during 1 to 11 days of gestation,0.4mg/kg/d of bromocriptine (dissolved in 75% ethanol) was injected subcutaneously during 6 to 8 days of gestation. The intervention group of pregnant rats was given Taishan Panshi San 10ml/kg/d during 1 to 11 days of gestation,0.4 mg/kg/d of bromocriptine were injected subcutaneously during 6 to 8 days of gestation. When the 12th day of gestation,20% urethane is used to anesthesia pregnant rats by intraperitoneal injection, when the pregnant rats had been anaesthetized and then opened the abdominal cavity. Blood serum samples from abdominal artery, serum levels of Interferon and Interleukin were measured by ELISA; Placentas and decidual tissue from pregnant rats were fixed in 4% paraformaldehyde, and then the expression of TNF-a in deciduas’ tissues of maternal-fetal interface were detected by immunohistochemistry.Results:1.The abortion rate of normal group, model group and intervention group were 0%,80%, 20%, and the abortion rate of model group was significantly higher than that of normal group and intervention group, the difference was statistically significant(P< 0.01, P<0.05).The difference between normal group and intervention group was not statistically significant(P> 0.05).2.The content of IFN-y in the serum of normal group, model group and intervention group were 9.15±3.01pg/ml,13.78±2.79 pg/ml,10.57±2.24 pg/ml, and the content of IFN-y in the serum of model group was significantly higher than that of normal group and intervention group, the difference was statistically significant(P<0.01, P<0.05). The difference between normal group and intervention group was not statistically significant(P>0.05).3. The content of IL-2 in the serum of normal group, model group and intervention group were 3.40±0.63 pg/ml,3.86±1.49 pg/ml,2.11±0.51 pg/ml, and the content of IL-2 in the serum of normal group and model group was significantly higher than that of intervention group, the difference was statistically significant(P<0.05,P<0.01). The difference between normal group and model group was not statistically significant(P>0.05)4. The content of IL-10 in the serum of normal group, model group and intervention group were 17.10±8.68 pg/ml,8.54±5.01 pg/ml,9.07±4.57 pg/ml, and the content of IL-10 in the serum of normal group was significantly higher than that of model group, the difference was statistically.significant(P< 0.05). The difference between model group and intervention group was not statistically significant(P>0.05).5.TNF-a of normal group and intervention group distribution difference in expression on side of the uterus were significantly lower than placenta in the maternal fetal interface, the difference was statistically significant(P<0.05).TNF-a of model group distribution difference in expression on sides of the uterus and placenta in the maternal fetal interface,the difference was not statistically significant(P>0.05).Conclusions:1. Bromocriptine induced abortion in pregnant rats, IFN-y high expression and IL-10 low expression in the serum of pregnant rats, TNF-α distribution abnormal in expression of the maternal fetal interface on both sides. It suggests that IFN-y, IL-10 and TNF-α may be related to the occurrence of abortion.2. Taishan Panshi San intervenes bromocriptine induced abortion has remarkable curative effect, it can reduce the content of IFN-y and IL-2 in the serum of abortion of rats, it also can improve TNF-a distribution in expression of the maternal fetal interface tends to the normal group. It suggests that Taishan Panshi San may through regulate the expression and distribution of IFN-γ and TNF-α, and then it can alter immune environment in vivo to avoid abortion.
