| ObjectiveTo investigate the protective effect of Xuanfei Tiaochang prescription on intestinal barrier of septic rats and the possible mechanism.MethodsSelect 36 SD male rats healthy male SD rats, weighing 200g±20g. Under standard breeding conditions for three days, they were randomly divided into six groups:sepsismodel group (LPS group, n=6), Xuanfei Tiaochang low dos group (XF low group, n=6), Xuanfei Tiaochang middle dose group (XF ingroup, n=6), Xuanfei Tiaochang high dose group (XF high group, n=6), Xuanbai Chengqi gas soup dose group(XB group, n=6) and control group(NS group, n=6). LPS intravenous injection modeling method:Before modeling, dissolve LPS with 0.9%sterile saline solutionat a concentration of 15mg/ml. Each rat is injected as lml/kg body weight. As a result, LPS intravenous injection was 15mg/kg body weight. Take blood specimens from the abdominal aorta and ileum tissue for relevant detection. Detection of indicators include:serum interleukin (IL-6) content; determination of serum malondialdehyde (MDA) and superoxide dismutase (SOD) activity and D-lactic acid levels;AMPK protein and p-AMPK protein expression was detected by western blot.Result1 inflammatory markersIL-6 and Serum MDAIL-6 and MDA in modle group and treatment groups were significantly higher than the normal group. The difference between the modle group and the normal group was statistically significant (P<0.05). Each treatment group was significant lower than the modle group. The difference between Xuanfei Tiaochang high dose group and Xuanfei Tiaochang middle dose group with the modle group was statistically significant (P<0.05).The IL-6 and MDA levels showed a decreasing relationship with the Xuanfei Tiaochang low, medium and high dose. The IL-6 and MDA levels in all the Xuanfei Tiaochang groups were lower than Xuanbai Chengqi group. The difference of the MDA levels between Xuanfei Tiaochang high dose group and Xuanbai Chengqi group was statistically significant (P<0.05).2 Oxidative stress indicators (Serum SOD activity)Serum SOD activity in modle group and the treatment groups were lower than the normal group. The difference between the modle group and the normal group was statistically significant (P<0.05). Each treatment group was higher than the modle group. The difference between Xuanfei Tiaochang high dose group with the modle group was statistically significant (P<0.05). The Serum SOD activity showed an increasing relationship with the Xuanfei Tiaochang low, medium and high dose. The Serum SOD activity in all the Xuanfei Tiaochang groups were powerful than Xuanbai Chengqi group, but the difference were not statistically significant (P>0.05).3 Serum D-lactic acidSerum D-lactic acid in modle group and treatment groups were higher than the normal group, the difference between the modle group and the normal group was statistically significant (P<0.05). Each treatment group was lower than the modle group. The difference between Xuanfei Tiaochang high dose group with the modle group was statistically significant (P<0.05). The Serum D-lactic acid levels showed a decreasing relationship with the Xuanfei Tiaochang loe, medium and high dose. The S Serum D-lactic acid in all the Xuanfei Tiaochang groups were powerful than Xuanbai Chengqi group, but the difference were not statistically significant (P>0.05).4 Morphological changes in intestinal tissueHistological changes of intestinal tissue of rats:The normal group rats have normal intestinal tissue, clear intestinal villi, complete mucosal surface and neat glands. But the villus of the modle group rats was congestion, edema, necrosis, and inflammatory cell infiltration visible intestine. All treatment groups’ rats have normal intestinal tissue, but the villus of every group exhibit varying degrees of congestion, edema, and inflammatory cell infiltration.5 AMPK protein and p-AMPK protein expression in intestinal tissueThe rats intestinal tissue AMPK protein expression in every groups showed no significant difference, but p-AMPK protein expression in every groups were statistically different. P-AMPK protein expression in modle group and the treatment groups were lower than the normal group. The difference between modle group with the normal group was statistically significant (P<0.05). Each treatment group was higher than the modle group. The difference between Xuanfei Tiaochang high dose group with the modle group was statistically significant (P<0.05). The p-AMPK protein expression showed an increasing relationship with the Xuanfei Tiaochang low, medium and high dose. The p-AMPK protein expression in all the Xuanfei Tiaochang groups was higher than Xuanbai Chengqi group, but the difference was not statistically significant (P>0.05).ConclusionObservation of the animal experiments showed Xuanfei Tiaochang can relieve sepsis intestinal tissue injury and improve bowel dysfunction. We think the possible mechanism may be related to the fact that Xuanfei Tiaochang can reduce sepsis oxidative stress and inflammatory response by activating AMPK pathway. |
