Objective: To observe the changes in the expression of p27KIP1 during the repair process of sciatic nerve injury treated with Buyang Huanwu Decoction(BHD).To explore the mechanism of Buyang Huanwu Decoction to promote peripheral nerve injury repair.Methods: The rats were randomly divided into control group,model group and BHD group.Model group and BHD group established sciatic nerve clamp injury model,control group did not perform modelling.After modelling,the BHD group was given gavage in the form of BHD,while the model and control groups were given saline gavage.The sciatic nerve function of the rats was examined by slant plate test and sciatic nerve function test at 3d,7d and 14 d.The sciatic nerve was taken 1h after gavage at 3d,7d and 14 d,and the morphological changes were examined by HE staining;the expression of p27KIP1 was examined by immunoblotting and immunohistochemistry.Schwann cells RSC96 were purchased,and cell proliferation was detected by CCK-8 after intervention with BHD-containing serum,and p27KIP1 expression was detected using immunofluorescence.Results: The results of the slant plate test and the neurological function index test showed that the scores of the BHD group were higher than those of the model group,and the scores gradually increased at 7d and 14 d compared to 3d.The HE staining results showed that the nerve morphology of the BHD group was better than that of the model group at 3d.At 7d and 14 d,the tissue arrangement of the model group and the BHD group gradually became regular and the degree of disorder gradually recovered,and the SCs gradually decreased,and the BHD group improved more than the model group.The degree of improvement was better in the BHD group than in the model group.The results of immunohistochemistry and immunoblotting showed that on day 3,the expression of p27KIP1 in the sciatic nerve of the BHD group decreased compared to that of the model and control groups;there was no significant difference in the expression of p27KIP1 in the model group compared to the control group.At 7d,the expression of p27KIP1 decreased in both the model and BHD groups compared to the control group;the expression of p27KIP1 was also significantly lower in the BHD group compared to the model group.At 14 days,the expression of p27KIP1 was significantly decreased in both the model group and the BHD group compared to the control group,and there was no significant difference in the expression of p27KIP1 in the BHD group compared to the model group.Immunofluorescence results showed that the expression of p27KIP1 was reduced in the blank serum group and the BHD-containing serum group compared to the control group.The expression of p27KIP1 was reduced in the BHD-containing serum group compared to the blank serum group.Conclusion: 1 Buyang Huanwu Decoction can improve nerve function in a model of sciatic nerve injury.2 Buyang Huanwu Decoction can improve the pathological morphology of the sciatic nerve injury model rats.3 Buyang Huanwu Decoction may promote the repair of sciatic nerve injury by reducing the expression of p27KIP1 protein to promote the proliferation of SCs. |
