Objectives:it is to investigate the effect of serum lipid on atherosclerosis(AS) in rats, by observing the effects of serum lipid on leptin and adiponectin, to investigate the mechanism of protecting vascular endothelium and anti atherosclerosis.Methods:36 SD rats were randomly divided:6 rats in the normal control group(Group A),10 rats in the model group(Group B),10 rats in the serum lipid group(Group C),10 rats in the rosuvastatin group(Group D). They were fed in different cages and kept with the constant temperature of 25 degree Celsius. In the 12 weeks of the modeling stage, the rats in the normal control group were given ordinary foods and free drinking water treatment;The rest of the group, at the beginning of the experiment, under aseptic operation, the model was made by intraperitoneal injection of vitamin D3 600 thousand U/lkg. At the end of fourth weeks, the model was made by intraperitoneal injection of vitamin D3 100 thousand U/lkg again. Usually they were to be fed with high fat diet and free drinking water. At the end of the 12 weeks, from the Group B, C and D randomly,2 rats were chosen killed in each group. Pathological staining was performed in their arteries. AS plaque formation was observed under microscope, and it was to confirm the success of AS model. Then it was enter the stage of treatment. During the 4 weeks of treatment, normal control group was administered with the same volume of distilled water (2ml), were given ordinary foods. The same volume of distilled water (2ml) in the model group was administered with the same volume of the medicine group, were to be fed with high fat diet. Serum lipid group received 8.19g/kg D (2ml) by intragastric administration of serum lipid, the rosuvastatin group received dose of 1.05mg/kg D (2ml) of rosuvastatin gavage, continuing to be fed with high fat diet, free drinking water. After 16 weeks, fasting for 12 hours, blood preparation and examination of the inferior vena cava after intraperitoneal anesthesia, and preparation and examination of thoracic aorta. ELISA method was used to detect the levels of leptin and adiponectin in rats. The pathological section of thoracic aorta was to observe the pathological changes of atherosclerosis.Results:after 16 weeks of treatment, in serum lipid group, leptin, adiponectin and model group were statistically significant (P<0.05) before and after treatment. There was no difference among the serum leptin, adiponectin in serum lipids and serum lipids (P>0.05). The pathological sections of thoracic aorta in the blood fat group showed that the pathological changes were significantly lower than those in the model group. Serum lipid and rosuvastatin both effectively controlled the formation of atherosclerotic lesion in rats.Conclusion:Serum lipids can reduce leptin by increasing adiponectin, and effectively reduce the degree of atherosclerotic lesions. So it can be proved that serum lipid can achieve the therapeutic effect of prevention and treatment of atherosclerosis.
