Objective:To observe the Zhike Gancao Decotion on the rat lumbar intervertebral disc model transplanted nucleus pulposus tissue PLA2 activity,IL-6 and TNF-a expression and the express ion of MMP-3 and TIMP-1 protein expression to explore the Zhike Gancao Decotion on reduce rat lumbar disc hermiation model of inflammation and prevent the degeneration of interverteral disc.Methods:The 90 SD male rats using random number table method, were randomly divided into for three groups, with 30 rats in each group, model group, TCM group (Zhike Gancao Decoction ), western medicine group (Indomethacin).TCM group:Zhike Gancao Decoction 9.1ml/kg.Befo-re using the drug 5 days,1 times/day and before modeling 1 hour gavage administration; wester n medicine group:indomethacion 7.5 mg/kg in the medication before modeling 1 hour gavage administration; model group according with the same dose and method of TCM group to medici-ne.At 7,14 and 28 days after modeling, the nucleus pulposus was removed and the pathological changes of the nucleus pulposus were obserned;activity values in the transplanted nucleus pulpo-sus tissue changes of PLA2;the expression of IL-6 and TNF-a were observed by immunohistoch-emical method.Using WESTERN-blot method-detect transplanted within the nucleus pulposus MMP-3 and TIMP-1 protein expression.Results:①By pathological HE staining analysis, Zhike Gancao Decoction group from modeling after 7,14 and 28 days of nucleus pulposus histopathological observation of nucleus pulposus and fiber ring exists in a few new blood vessel growth, infiltration of inflammatory cells, nucleus cytoplasm hole like change, around nucleus pulposus cartilage formation evaluation results than model group at the same time, with different degrees of change. ②After seventh days, theactivity of PLA2 in the experimental group reached the highest peak, but the TCM group and Western me dicine group were significantly lower than the model group. On the seventh day, the PLA2 activit-y of the TCM group was significantly higher than that of the western medicine group, the TCM group and the model group were compared with P<0.05, the western medicine group and the m-odel group P<0.01; On the 7th day, and the influence of PLA2 activity in the nucleus pulposus in the TCM group was better than western medicine group obviously, between chinese and western Medicine (P> 0.05), no significant statistical difference that 7 days in the two groups can be the nucleus pulposus of PLA2 activity is inhibited by, but TCM group is better than western medicin-e group was obvious. At 14 and 28 days, chinese and western medicines are groups of PLA2 acti-vity in the nucleus pulposus play an inhibitory effect (P<0.05), model group, the nuclleus pulp-osus of PLA2 activityvalues are still at a high level.In each time period, chinese and western two group effect in the treatment without significant difference, Zhike Gancao Decoction can also inhibit inflammatory cytok-ines cytokines and reduce the role of PLA2 activity in nucleus pu-lposus.③In 28 days after modeling, the transplantation of nucleus pulposus cells were results of corresponding parts were yellow or brown stain.Model group IL-6 and TNF-α expression and higher than western medicine group and TCM group;IL-6 and TNF-α expression medicines exp-ression than western medicine group decreased.In the 28 days after modeling, transplanted nucleus of IL-6 and TNF-α expression in western me-dicine group and the model group, the difference was statistically significant (P<0.05), the TCM group and the model group, the difference was statistically significant (P<0.05);The TCM and western drugs P> 0.05 not statistically different, indicating that the drug reduced the nucleus in the West there is no difference on the IL-6 and TNF-α expression. From the statistics can be seen in the TCM group of transplantation of nucleus pulposus in IL-6 and TNF-α expression was sig-nificantly better than the western medicine group, indicating Zhike Gancao Decoction can inhibit the expression of nucleus pulposus of IL-6 and TNF-α, and delay the degeneration of interverteb-ral disc. ④In the 28 days after modeling, transplanted nucleus pulposus of MMP-3 and TIMP-1 protein expression in Western medicine group and the model group, the difference was statistica-ally significant (P<0.05), TCM group and the model group, the difference was statistically sign-ificant (P<0.05);As can be seen from a statistically TCM group for MMP-3 and TIMP-1 protein expression in the transplanted nucleus significantly better than western medicine group. So that the Zhike Gancao Decoction can inhibit the expression of MMP-3 protein in the nucleus pulpos-s, thus delaying the degeneration of intervertebral disc.Conclusion:①The Zhike Gancao Decoction can improved inflammatory mediators in the perip-heral transplantation of nucleus pulposus and annulus fibrosus gathered in histopathology;②The Zhike Gancao Decoction can reduce the role of PLA2 in the nucleus pulposus tissue to a certain extent, and has the same effect as anti inflammation and analgesic effect;③The Zhike Gancao D-ecoction can inhibit the expression of IL-6 and TNF-α in the positive cells of the nucleus pulpos-us, inhibit the synthesis of inflammatory mediators, through down regulating the protein expres-sion of MMPs and TIMP-1, and play a role in delaying the degeneration of intervertebral disc.
