Background:Endometriosis (EMs) is a common gynecological disorder characterized by the presence of endometrial glands and stroma outside of the uterine cavity, which associated with chronic abdominal pain, dysmenorrhea, and infertility and so on. However, the pathogenesis is controversial. Although EMs is a benign disease, it has great similarity with cancer in the invasion, angiogenesis, proliferation, apoptosis and many other aspects. Previous study has confirmed that fragments of the endometrium survive, grow and cause symptoms by adhesion, invasion and angiogenesis. Ezrin, as a cell membrane-cytoskeletal protein, is involved in cell adhesion and the formation of cell structure. Additionally, Ezrin is PKA anchoring protein. NHERF1, aslo knowed as Ezrin-radixin-moesin binding phosphoprotein 50, play an important role in tumor cells adhesion, migration and other processes. Thus, activation of Ezrin combined with NHERF1 may be involved in cell adhesion and migration mechanisms of EMs by cAMP-PKA pathway.Objective:In this study, we designed to look at the expresstion of Phospho-Ezrin, NHERF1, PKAⅠα in ectopic endometrial tissue and eutopic endometrial tissue in women with EMs and endometrial tissue in non-EMs patients.Materials and Methods:We obtained ovarian endometriomas and endometrial tissues from patients with stage Ⅲ-Ⅳ EMs (according to revised AFS classification). And the endometrial tissues from patients without EMs under hysteroscopy and laparoscopy due to infertility or septate uterus during the same period rolled in control group. We looked at the expression of Ezrin, NHERF1, PKAⅠα mRNA by real-time RT-PCR; and used Western blot and immunohistochemistry to detect the location and semi-quantitative analysis of Phospho-Ezrin, Ezrin, NHERF1, PKA I ain different endometriums.Result:1. Real-time RT-PCR results:The expression of PKAⅠα mRNA:ectopic endometrium> eutopic endometrium> control group, there were statistically significant differences between groups (P<0.05); However, the expression of Ezrin, NHERF1 mRNA in the three groups had no significant differences(P>0.05).2. Western blot results:The expression levers of Phospho-Ezrin and PKAⅠα: ectopic endometrium> eutopic endometrium> control group, there were statistically significant differences between groups (P<0.05); However, the expression of Ezrin, NHERF1 in the three groups had no significant differences(P>0.05).3. Immunohistochemistry results:Phospho-Ezrin, Ezrin, NHERF1, PKAⅠα in EMs ectopic endometrium, eutopic tissue and normal endometrium were expressed, mainly in the endometrial glands and glandular epithelium on the cell membrane and cytoplasm. The expression levers of Phospho-Ezrin and PKAI a:ectopic endometrium> eutopic endometrium> control group, there were statistically significant differences between groups (P<0.05); However, the expression of Ezrin, NHERF1 in the three groups had no significant differences.Conclusion:1. The expression of Phospho-Ezrin, PKA I a:ectopic endometrium> eutopic endometrium> control group, there were statistically significant differences between groups (P<0.05).2. Phospho-Ezrin and PKA-mediated signal pathway may be involved in the pathogenesis of endometriosis.
