Objective:Mainly through observing the dry eye patients’s symptoms before and after the treatment about tear secretion, break-up time, sodium fluorescein corneal staining, conjunctival impression cytology, thus to evaluate Xiao Shengsan granules’efficacy,and explore its influence about goblet cells, then though combined with animal experiments, conduct further verification and discussion.Methods:1.The 30 cases (60 eyes) were randomly divided into two groups.Group A is control group:use sodium hyaluronate eye drops,four times a day,every time a drop.Group B is TCM group:use granules of xiaoshengsan, it’s mainly composed of granules is baishao, chaihu, renshen,danggui,wuweizi,maidong, ect.twice a day,every time a bag.Observe the score of symptoms,break up time(BUT),schimer(SIT),fluorescein staining(FL),impression cytology(IC) in tear respectively respectively before and after the treatment, one week、two weeks、three weeks and four weeks after the treatment, At the same time measured the amount of symptoms score、BUT、SIT、FL、IC.2.180 male C57BL/6 mice were divided into six groups, Group A:normal; Group B: subcutaneously intramuscularly scopolamine; Group C:subcutaneously intramuscularly scopolamine+physiological saline lavage; Group D:subcutaneously intramuscularly scopolamine+granules of low xiaoshengsan lavage; Group E:subcutaneously intramuscularly scopolamine+granules of middle xiaoshengsan lavage; Group D: subcutaneously intramuscularly scopolamine+granules of high xiaoshengsan lavage.Then determined the SIT、BUT、SIT、FL、IC respectively before and after subcutaneous injection the scopolamine, one week、two weeks、three weeks and four weeks after the treatment.Results:1.After the treatment, Group B significantly improved in symptoms,and BUT、SI、the number and grade of goblet cell and FL, and had statistically significant differences(p <0.05).Group A only had good effects on symptoms and FL, and the number of goblet cell, they all had statistically significant difference (p<0.05), but had no significant difference about SIT、BUT、the number of goblet cell(p> 0.05).2.The BUT and SIT、FL、IC of group B, C, D, E,F was significantly change after subcutaneously intramuscularly scopolamine (p<0.05), while group A was not significantly different (p> 0.05).The BUT. SIT. FL. the number and grade of goblet cell of Group E、F was chananged and had a significant difference comparing subcutaneously intramuscularly scopolamine (p<0.05), The BUT. FL of Group D was chananged and had a significant difference comparing subcutaneously intramuscularly scopolamine (p<0.05),while the SIT、 the number and grade of goblet cell had no significant (p>0.05). The BUT. SIT. FL. CI of Group B. C was not chananged and had no significant(p>0.05). while group A was not have significantly different (p> 0.05).Conclusion:The granules of xiaoshengsan can relieve the score of symptoms.and increase the tear secretion, reduce the damageand of goblet cell,elevate tear film stability,reduce ocular surface damage.Its mechanism may be by regulating the immune system, reduce the intensity of the inflammatory response, reduce the release of pro-inflammatory cytokines, affecting the ocular surface epithelial cells, lymphocytes and apoptosis of lacrimal gland, and to prevent the goblet cells apoptosis, thus play its pharmacological effects, then inhibit or delay the formation of dry eye.
