Objective:By observing the subjects before and after the treatment of tear film breakup time(BUT), tear film interferometry and cytokine TGF-β in the tear. Dry eye patients with venous blood sex hormone level, evaluated the efficacy of Granules of Xiaoshengsan, investigated its role of immune-mediated inflammatory in molecular level combined with the animal experiments.Methods:1.30cases(60eyes) were randomly divided into two groups, Group A-control group:artificial tears every time a drop three times a day; Group B-TCM group:granules of Xiaoshengsan, a bag twice a day. Observed the tear film breakup time (BUT) and Schirmer1at two weeks and four weeks after treatment.2. ELISA before and after treatment were detected TGF-beta factor content in tears.3. Patients with dry eye phlebotomize on an empty stomach, detection of sex hormone.4.80BALB/c male mices were divided into six groups, Group A: low-level; Group B:mid-level; Group C:high-level; Group D:Physiological saline;Group E:model;Group F:normal.Measured Schirmer1two weeks and four weeks after treatment. Then do HE, immunohistochemistry. Results:1. After treatment, Group B improved significantly in BUT, SIT1and had statistically significant differences(p<0.05).2. The content of TGF-β of Group B in tear increased after treatment, but no significantly different(P>0.05).3.30cases are detected, sex hormone abnormal13cases, testosterone (T) reduced as the main abnormal indicators, including5cases (38.46%).4. The SIT of group A、B、C、D、E were significantly shorter after building comparing preoperative(p<0.05).The SIT of Group A、B、C were lengthened in28days observation period and had a significant difference comparing postoperative while other groups had no significant (p>0.05). The expression of IL-1and IL-6in mice cornea significantly decreased after treatment(p<0.05). Conclusion:Granules of Xiaoshengsan can increase the tear secretion, elevate tear film stability and increase the content of TGF-β in tear. Its mechanism may be mainly related to intervention inflammatory cytokines, reducing proinflammatory cytokine release. Dry eye associated with testosterone decrease.
