Objective:The study was accorded to the Atherosclerosis model by SD rates, combining HE staining, imumunohistochemistry, polymerase chain reaction(RT-PCR) to investigate the expression of TLR6 and the effect of rosuvastatin on TLR6 and atherosclerosis of atherosclerotic rat aorta.Method:Prepare 36 healthy male SD rats which weight between 180g-200 g, then divided into control group(n =12) and experimental group(n=24) randomly after one week adaptive feed. Rats in the experimental group were high cholesterol fed for 4 weeks, then randomly divided into two groups: model group(n=12) and rosuvastatin group(n=12). NS and rosuvastatin(5mg/kg/d) were prescribed separately for 4 weeks. At the end of experimental period, the serum levels of TC, TG, LDL-C and HDL-C were detected. The pathological changes of aorta was evaluated by HE staining. The expression of TLR6 of aorta was observed by immunohistochemical techniques. The expression of TLR6 m RNA of aorta was measured by reverse transcription polymerase chain reaction(RT-PCR).Results:1.Blood lipid: The levels of TC and LDL-C in model group and rosuvastatin group were both elevated compared with control group(P<0.05). The TC and LDL-C levels in rosuvastatin group were significantly lower than the model group(P<0.05). There were no significant differences of TG and HDL-C level among the three groups(P>0.05).2.Analyses of Histological and Morphology: The thoracic aortic wall structure of each layer was relatively intact, smooth intima, no lipid calcification and deposition were observed in control group. In model group, intima obviously thickening, a lot of smooth muscle cells hyperplasia under the intima, a lot of foam cells to gathered, a large number of lipid deposition and formation of atherosclerosis were observed. A significant regression of atherosclerosis changes was found in rosuvasatin group when compared with the model group.3.The results of Immunohistochemincal: In all thoracic aorta, there were TLR6 expression. A small amount expression of TLR6 was observed in aorta in control group. An increase of TLR6 was observed in both model group and rosuvasatin group when compared with control group(P<0.05). A decrease of TLR6 was found in rosuvastatin group as compared with model group(P<0.05).4.The results of fluorescence quantitative PCR: In model group, an increase of the expression of TLR6 m RNA was observed compared with control group and rosuvastatin group. In rosuvastatin group, a decrease of the expression of TLR6 m RNA was found when compared with model group.In control group, the expression of TLR6 m RNA was less and scttered. The expression of TLR6 m RNA in each group have a significantly differents(P<0.05).Conclusion:1. The expression of TLR6 in atherosclerotic rats increased.2. Rosuvastatin may place a role to improve atherosclerosis by reducing the expression of TLR6.
