| Objective:To study the expression of FBXL20 in hepatic carcinoma.Method:lmmunohistochemical SP method was used to detect the expression of FBXL20 in 113 liver tissue from 5 different disease(hepatocellular carcinoma, fatty liver, hepatitis B, hepatitis C and autoimmune liver disease).4 hepatoma cell lines (SMMC-7721, HepG2, HepG2.2.1.5, Huh7) and normal liver cells L02 cells were cultured. The expression of FBXL20 was analyzed by Western blot.Results1. In hepatocellular carcinoma, the positive rate of FBXL20 was 90.9%(60/66), and that was 57.6%(38/66) in para-cancerous tissues. And in hepatocellular carcinoma, the strongly expression rate of FBXL20 was 652%(43/66). Expression of FBXL20 protein was higher in hepatocellular carcinoma tissues than that in para-cancerous tissues (P<0.05). The expression of FBXL20 was correlated with pathological grade (P<0.05), and there was no relationship among age, sex, tumor sides, AJCC TNM stage, HBsAg and liver cirrhosis. Expression of FBXL20 protein was higher in hepatitis B and hepatitis C tissues than that in Fatty liver tissues (P<0.05). And the positive rate of FBXL20 in hepatocellular carcinoma was significantly higher than that in other hepatitis (P<0.05).2. The expression of FBXL20 protein in normal human hepatocytes was at a certain degree, and the expression of FBXL20 increased in HCC cell line SMMC-7721 (the ratio was 1.056±0.167 vs. 0.775±0.048, P<0.05). There was no significant in FBXL20 expression between HepG2 and L02 (the ratio was 0.701±0.028 vs. 0.775±0.048, P>0.05). And in the HepG2.2.1.5 and Huh7 the FBXL20 protein expression level was lower than that of L02 (the ratio was 0.487±0.052 and 0.034±0.096 respectively, vs. 0.775±0.048, P<0.05).Conclusion1. FBXL20 over-expressed in hepatocellular carcinoma. The strongly expression of FBXL20 was correlated with pathological grade.2. Expression of FBXL20 protein was higher in hepatitis B and hepatitis C tissues than that in fatty liver tissues (P<0.05). And the positive rate of FBXL20 in hepatocellular carcinoma was significantly higher than that in other hepatitis.3. The expression of FBXL20 protein in normal human hepatocytes was at a certain degree, and the expression of FBXL20 increased in HCC cell line SMMC-7721. And the FBXL20 protein expression level decreased in the HepG2.2.1.5 and Huh7.It may be involved in the carcinogenesis of hepatocellular carcinoma. |
