Objective:To investigate the expression of FBXL20 in esophageal cancer,and to explore its possible role and potential mechanism in esophageal cancer.In order to provide a new biomarker for esophageal cancer can be cured completely in the future.Methods:To evaluate FBXL20 expression in esophageal cancer tissues and cancer adjacent tissues,immunohistochemistry was performed.The FBXL20 expression in five esophageal cancer cell lines was also explored by using q RT-PCR.The sh RNA lentivirus and vector were transduced into relatively high cells.The expression of FBXL20 m RNA and protein in esophageal cancer cells after transfection was analyzed by q RT-PCR and Western blot.We further determined the effects of FBXL20 on esophageal cancer cell cycle and apoptosis using flow cytometry.To investigate the effect of FBXL20 on esophageal cancer cell growth,CCK-8 assays and colony formation were used.In order to validate the effects of FBXL20 on migration and invasion ability in esophageal cancer cell,the transwell assay was performed.To investigate the potential mechanism of FBXL20 in promoting esophageal cancer cell cycle and metastasis,the relationship between FBXL20 and the Wnt/β-catenin signaling pathway was explored.Statistical analysis was performed using Graph Pad Prism5 and SPSS17.0.Results:The present study demonstrated that the expression of FBXL20 in esophageal cancer tissue was significantly higher than its expression in adjacent tissues.We demonstrated that FBXL20 was an oncogenic gene in the esophageal cancer.In vitro studies,silencing FBXL20 expression by lentiviral vector sh RNA induced G0/G1 cell cycle arrest,and inhibited the proliferation,migration and invasion ability of esophageal cancer cells through Wnt/β-catenin signaling pathway.In addition,FBXL20 silencing could increase the apoptosis of esophageal cancer cells.Conclusion:This study demonstrated that FBXL20 was an oncogenic gene.FBXL20 might serve as a potential tumor marker and therapeutic target for esophageal cancer.We have explained the Potential mechanism of FBXL20,which may provide additional methods and strategies for the treatment of patients with esophageal cancer.
