Objective:We mainly study the preventive effect of Saponions of panax ginseng and Ginsenoside Rb1 on the diabetes melitus and the dose-effect relationship between Saponions of panax ginseng and the diabetes mellitus, we analyze deeply that the interaction of Ginseng saponin monomer in Saponions of panax ginseng on diabetes mellitus is better than Ginsenoside Rbl,in order to provide a theoretical basis for the prevention of the diabetes mellitus to achieve the primary prevention.Methods:We divided the 21 healthy and healthy female SD rats into 4 groups randomly, Group A:model groups(6 rats); Group B:low-dose Saponions of panax ginseng (40mg/Kg) group(5 rats); Group C:high-dose Saponions of panax ginseng (80mg/kg) group(5 rats); Group D:high-dose Saponions of panax ginseng (80mg/Kg) and GS-Rb1 (15mg/kg) group(5rats). Group B-. C rats were injected into different doses (20,40 mg/Kg) Saponions of panax ginseng solution respectively. Group D rats were injected into high-dose Saponions of panax ginseng (80mg/kg) and GS-Rbl (15mg/kg) group(5rats); Group A rats were injected the same volume of distilled water by the same method. Rats were injected 5 times on 10 days. We draw the blood to measure the FPG on the second day. Rats were injected 1%STZ solution on the 13th days. We observe the variation of the weight and the food and water intake 12 days continuously. We got the blood and pancreas, kidney and liver samples. We divided the serum into two parts, one part were given the Yanbian Hospital to test the TC、TG、ALT、AST、 BUN、Scr of the serum, one part were measure the SOD and MDA in our own laboratory. The pancreas, kidney and liver samples were placed in light microscope to observe the pathological changes after HE staining.Results:Compared with the three other groups, the food and water intake of Group A rats increased faster. Compared with Group C and Group D, the food and water intake of Group B rats increased relatively faster. The food and water intake of Group C and D increased equally. Compared with the three other groups, the body weight growth rate of Group A rats increased slowest dramaticly (P<0.05), Compared with Group B, the body weight growth rate of Group C and Group D increased faster dramaticly (P<0.05), but the body weight growth rate of Group C and D increased equally(P>0.05). Compared with Group A,the glucose level of Group B, Group C and Group D all declined dramaticly (P<0.05),compared with Group B, the glucose level of Group C and Group D also declined dramaticly (P<0.05),but the glucose level of Group C and D declined equally (P>0.05). Compared with the three other groups, the TC and TG level of Group A rats increased dramaticly(P<0.05), Compared with Group B, the TC and TG level of Group C and Group D declined dramaticly(P<0.05), but the TC and TG level of Group C and D declined equally(P>0.05).Compared with the three other groups, the ALT and AST level of Group A rats increased dramaticly (P<0.05), Compared with Group B, the ALT and AST level of Group C and Group D declined dramaticly (P<0.05), but the ALT and AST level of Group C and D declined equally(P>0.05). Compared with Group A, the SOD level of Group B, Group C and Group D increased dramaticly,but the MDA level declined dramaticly (P<0.05). Compared with Group C and Group D,the MDA and SOD level of Group B have statistical differences (P<0.05),but the SOD and MDA level of Group C and D were equal (P>0.05). Compared with the three other groups, the BUN and Scr level of Group A rats increased dramaticly (P<0.05), Compared with Group B, the BUN and Scr level of Group C and Group D declined dramaticly(P<0.05), but the BUN and Scr level of Group C and D declined equally(P>0.05). Pancreas organization pathology shows that pancreas islet of group A has compensatory hypertrophy. There is Irregular strip break in positive area and Coloring shallow, loose arrangement. The boundary of pancreas is not clear; compared with group A, compensatory hypertrophy of Group B, Group C and Group D is lighter, the color of positive area is deeper, pathology of some rats islet cells is close to normal. Compared with group B, the pancreas damage of Group C and Group D is obvious lighter. Kidney organization pathology shows that glomerulus volume of Group A rats is significant bigger with inflammatory cell infiltration, more mesangial matrix and basilar membrane becoming thicker. The blood capillary in the glomerulus become smaller, the epithelial cells of some kidney tubules arrange loosely with slight edema. Compared with group A, the kidney damage of Group B is obvious lighter. The glomerulus volume become obvious smaller, the phenomenon of inflammatory cell infiltration is lighter. The pathology of kidney tubules and renal interstitium is not obvious. The blood capillary of basilar membrane and mesangial area is close to normal. Compared with group B, The Kidney damage of Group C and Group D is lighter. Liver organization pathology shows that structure of hepatic cords is disorder. The hepatic sinusoid become smaller or disappeared. hepatocyte become bigger with varying degrees and diffuse fat degeneration, a minority of cells shows vacuolar degeneration. Compared with group A, structure of hepatic cords is regular, vacuolar degeneration of hepatocyte disappears. The phenomenon of fat degeneration become lighter. Compared with group B, the hepatic sinusoid of Group C and Group D is almost close to normal, structure of hepatic cords is regular, the phenomenon of fat degeneration become lighter. We can see hepatic lobule.Conclusion:1.Saponions of panax ginseng has a preventive effect on diabetes mellitus rats; within limits, Increasing the Saponions of panax ginseng dose can improve the preventive effect,there is no internal physical damage.2. The preventive effect of Saponions of panax ginseng is better than GS-Rb1,the phenomenon is relative with the creative and synergistic effect of Ginseng saponin monomer in Saponions of panax ginseng.3.The preventive effect mechanism of Saponions of panax ginseng and GS-Rbl is relative with ginseng antioxygenation that delince the MDA level and increase the SOD level to reduce the ROS level,in order to avoid other internal physical damage.
