Objective:To investigate the lncRNA expression and regulation network in postmenopausal osteoporosis with Kidney Yin deficiency.Methods:Human IncRNA expression microarrays were applied to explore gene expression spectrum’s difference. By TCM syndrome, patients with postmenopausal osteoporosis were randomly divided into three groups:kidney Yin deficiency, kidney Yang deficiency, and healthy postmenopausal women also were selected as control group. Kidney Yin deficiency group was compared with other two groups respectively.The genes with common significant difference were studied by bioinfomation analysis. Get the biological functions and the biological processes by GO analysis and pathway analysis. Real-time RT-PCR was used to examine the xpression of LINC00334, LINC00189, LOC101929378 and the results of microarray were confirmed.Results:1.Compared with kidney Yang deficiency group, there were 35 mRNAs and 45 lncRNAs expressed differentially in the kidney Yin deficiency group. Compared with control group, there were 1867 mRNAs and1926 lncRNAs expressed differentially in the kidney Yin deficiency group.Compared with control group, there were 8 lncRNAs (LINC00334、 LINC00189、 LOC101929378、 XLOC01392、 ENSG00000237489.2、 ENSG00000225278.2 、AK125437 and RNA95672)expressed differentially in the kidney Yin deficiency group.2. Pathway analysis indicated that 12 potential pathways corresponded to these aberrant lncRNAs.3.LINC00189 was frequently down-regulated in the kidney Yin deficiency group(FC=4.71, P=0.007). LINC00334(FC=6.83, P=0.005) and LOC101929378 were up-regulated in the kidney Yin deficiency group(FC=4.51, P=0.035).The change trend was coincident with the result of CHIP.Conclusion:There was some association between postmenopausal osteoporosis with Kidney Yin deficiency and the expression of the lncRNAs LINC00334、 LINC00189、 LOC101929378 、 XLOC 013921、 ENSG00000237489.2、ENSG00000225278.2、 AK125437 and RNA95672.Jak/Stat,MAPK, Chemokine signaling pathway and Calcium signaling pathway were regulated by theose differentiated lncRNAs.
