ObjectiveUsing the technology of genechip to elucidate the characteristics of genes expression profiles of postmenopausal osteoporosis with Kidney Yin deficiency.Accordingly,the objective of the study is to find expressed differentially genes and to investigate biological information characteristics of related genes of postmenopausal osteoporosis with Kidney Yin deficiency.Methods10 patients with postmenopausal osteoporosis were randomly divided into three groups: Kidney Yin deficiency(n=4),Kidney Yang deficiency(n=3),non-Kidney deficiency(n=3), another 3 healthy postmenopausal women also were selected.Human gene expression microarrays were applied to explore gene expression spectrum's difference of the groups. Kidney Yin deficiency group was compared with other three groups respectively. The genes with common significant difference were studied by Go and Pathway analysis. According tc the results of genechip analysis, genes expressed differentially of Kidney yin deficiency and Healthy group were randomly selected for realtime PCR(RT-PCR) validation.Results1,There is characteristic profiles of differential expressed genes among Kidney Yin deficiency group with other three groups.The results showed 197 genes expressed differentially between Kidney Yin deficiency group and healthy group,among which 103 were up-regulated and 94 were down-regulated;354 genes between Kidney Yin deficiency group and Kidney Yang deficiency group,among which 275 were up-regulated and 79 were down-regulated;54 genes between Kidney Yin deficiency group and non-Kidney deficiency group,among which 13 were up-regulated and 41 were down-regulated.9 genes were expressed differentially among Kidney Yin deficiency group with other three groups: PROK2,ASB1,MUC12,GSTM5,CLCF1,GPR27,C3 or F35 and urknown genes chr3: 113822408-113822347,chr18:34851804-34851745.Compared with healthy group,GSTM5 and MUC12 showed up-regulated expression,GPR27,C3orf35,ASB1,CLCF1 and PROK2 showed down-regulated expression.2,PROK2,ASB1,MUC12,GSTM5,CLCF1,GPR27 and C3orf35 in their analysis according to GO functions of the general classification.Mainly genes related to regulation of biological process,regulation of cellular metabolic process,signaling pathway,apoptosis, regulation of phosphate metabolic process,regulation of JAK-STAT cascade,immune system development,inflammatory response.3,The pathway analysis showed GSTM5 was involved in glutathione metabolism, Metabolism of xenobiotics by cytochrome P450 and Drug metabolism-cytochrome P450,and CLCF1 was involved in cytokine-cytokine receptor interaction and Jak-STAT signaling.4,The results of RT-PCR showed that,expressions of ASB1 and GSTM5 between the Kidney Yin Deficiency group and the normal group are consistent with results of the gene chips.ConclusionsThe pathogenesis of Postmenopausal osteoporosis with Kidney Yin deficiency is possibly associated with the genes expression of GPR27,ASB1,PROK2,CLCF1 and GSTM5,And it may correlated with glutathione metabolism pathway,cytokine-cytokine receptor interaction pathway and Jak-STAT signaling pathway.
