Objective Observe and compare the influence of different dog day moxibustion temperatures on the asthmatic New Zealand rabbits as well as the MMP-9 protein and mRNA expression in their lung tissues. Explore the correlation between pleural surface temperature and moxibustion on ginger as well as its relationship with the effect of’qi extending affected parts’.Methods Select 30 male New Zealand rabbits, body mass:(2.5±0.3)kg. Randomly divide them into five groups:blank control group, model control group, experimental group, control group and dexamethasone group. Each group has 6 New Zealand rabbits. Every group makes molds by using the asthmatic airway remodeling model except the blank control group. Model group only given airway remodeling, but not intervention.Experimental group and control group are given ginger moxibustion combined with the external application of Chinese medicine in dog days as intervention. Choose Dazhui and Feishu as acupoint. We use infrared cameras to monitor the temperature variation of two asthmatic rabbits pleural surface. Experimental group and control group are given intervention 15 times every other day. The pleural surface temperature of experimental group increases 1℃ compared to what’s before treatment. Control group is given three moxa cones. Dexamethasone group is given dexamethasone (0.5mg/kg) inhalation 30min. This is done every other day,15 times in all. Then, the right lungs of all the New Zealand rabbits are taken out for Immunohistochemistry to test MMP-9 protein expression. The left lungs are used for qPCR to test MMP-9 mRNA expression. Observe the change of MMP-9 in 5 groups, and analyze the correlation between pleural surface temperature and moxibustion on ginger as well as its relationship with the effect of ’qi extending affected parts’.Results 1.The MMP-9 protein and mRNA expression in the model group, model control group, control group, experimental group and dexamethasone group is significantly higher than that of the control group. The differences are statistically significant(P<0.05).The MMP-9 protein and mRNA expression in the control group, experimental group and dexamethasone group is significantly lower than that of the model control group. The differences are statistically significant(P< 0.05). The MMP-9 protein and mRNA expression in the experimental group is significantly lower than that of the control group. The differences are statistically significant(P< 0.05); The MMP-9 protein and mRNA expression in the experimental group and dexamethasone group has no significant difference, and the difference is not statistically significant(P> 0.05).2.The Pleural surface temperature of the experimental group after moxibustion is significantly higher than that before moxibustion, and the difference is statistically significant (P<0.05); The Pleural surface temperature of the control group has no significant difference between after and before moxibustion, and the difference is not statistically significant(P>0.05); After moxibustion, the surface temperature of the experimental group is significantly higher than that of the control group, and the difference is statistically significant (P<0.05); The Pleural surface temperature of the experimental group increases with the moxibustion on ginger of moxa cones. Spearman correlation coefficient is 0.433 and the difference is statistically significant (P<0.01).Conclusion When the moxibustion on ginger reaches nine moxa cones and nine moxa cones above,’qi extengding affected parts’might be induced by increasing asthmatic rabbits’ pleural surface temperature, which will reduce MMP-9 protein and mRNA expression and inhibit MMP-9 synthesis, improve asthmatic airway remodeling in rabbits, so as to effectively relieve asthma symptoms. At the same time, the surface temperature is positively correlated with the moxa cones of moxibustion on ginger. The elevated temperatures may be one of the indexes of’qi extengding affected parts’.
