Study On Preventing Obesity And Antilipemic Effects Of Haihe Jianfei Cream In Rats And Its Potential Mechanism

Objective:Study on the mechanisms of Haihe Jianfei cream on preventing obesity and antilipemic in rats.Methods:Female and male SD rats (60±5 g) after borning 24 days, were chosed and randomly devided into 6 groups as Control group, Model group, Haihe jianfei cream high dose group(1.08 g/kg/d), Middle dose group(0.36 g/kg/d), Low dose group(0.18 g/kg/d), Orlistat positive drug group(60 g/kg/d). Control group and Model group were given the same volume physiological saline. All groups were given high fat diet excepted Control group which was fed with normal diet. The groups were continuously gavaged administration for 8 weeks, and the general state of health, four blood serum indexes including cholesterol(TC),triglyeerides(TG), high-density lipoprotein(HDL), low-density lipoprotein(LDL), the weight of liver, the wet weight of body fat and Lee’s indexes of rats were measured and recorded during the experiment. The pathological structural variation of liver cell and adipose cell, the sizes and number of fat cells were observed by HE staining technique. The LEP, TNF-α, CORT and E2 level in serum were investigated by ELISA.Results:Compared the model group with the Control group, it is shown that the body weight, body fat, TC, TG, HDL, LDL, LEP, TNF-α, CORT and E2 level in serum were increased.The liver cells were appeared patholo-gical structure, the sizes of fat cells were significantly bigger and lots of lipid droplets were found in the adipose cells. Female rats in Haihe jianfei cream group showed decreasing in the body weight and body fat coefficient, but there was no significant differences in the mearsured indexes on male rats. The level of TC, TG, LDL were lower than modle groups in blood biochemical indexes analysis without sex difference. Haihe jianfei cream in three dose level all had high improvement, especially for the female rats, in ameliorating the structure of liver cell, prohibiting the aggregation of lipid droplets and decreasing the size of the fat cells. Haihe jianfei cream could also decreased the level of LEP, TNF-a, CORT and E2 in the blood of female rats which was detected by ELISA, however, in the blood of male rats, the level of LEP and TNF-a unconspicuously decreased, and the level of CORT was significantly lower than Control group, while the level of E2 was much higher than Model group.Conclusion:This research showed that Haihe Jianfei Cream could decrease lipidrising and obesity on female rats.For the male rats, the cream could prevent lipidrising but no weight loss effect. The explanations for the gender diversity on the observations above were probably the differences on the level of leptin, TNF-α, glucocorticoids and estrogen.
