| In this work,extraction and purification of fucoxanthin from Lamiannia Japanica was studied,moreover,the stability of fucoxanthin was also researched.First, it was researched that optimization and comparion of convention solvent extraction(CSE) and ultrasound assisted extraction(UAE) of fucoxanthin from Laminania Japanica using response surface methodology. Three independent variables such as extraction temperature (℃), ratio of solvent to raw material and extraction time (min) were investigated. The second-order polymial equation predicted the extraction condition for the highest yield at 123 min extraction time,53.42℃extraction temperature, 43.53 (mL/g) liquid-solid ratio.The developed model predicted fucoxanthin yield of 0.23049mg/g at the optimal conditions(CSE) and 60.99 min extraction time,50.13℃extraction temperature,33.52 (mL/g) liquid-solid ratio.The developed model predicted fucoxanthin yield of 0.348mg/g at the optimal conditions(UAE). the average fucoxanthin content was more than51.19% and solvent depletion and time reduce 22.1% and 50.14%,respectively Second, Crude fucoxanthin extracts were purified by Low-pressure column chromatography (LPCC) with chloroform and methanol as eluent. Compared with the results between isocratic elution and gradient elution, it was better that purified fucoxanthin product was prepared by gradient elution with chloroform and methanol as eluent. The gradient elution was performed by A and B solvent systems, from 0 to 100%B in 100 min., the purities of the fucoxanthin product was 91.6% and recovery of product was 92.1%.Third, the feasibility of recovering solvent and concentration purication from methanol solution of fucoxanthin by Nanofiltration (NF) was studied. Five commercially available polymeric NF membranes were employed and STARMEMTM 122 showed acceptable flux and high retention.Last,the stability of fucoxanthin was system researched.The influences of Light,temperature,antioxidant and PH was analytical investigation which supply the theory foundation for the extraction procedure and preservation of fucoxanthin. |
