| Studies show that protamine(Protamines, PRM) have great correlation with male infertility, which is the main protein binding and DNA sperm nucleus. The latest reports show that PRM1 is a form of PRM, which may be associated with the pathogenesis of gastric adenocarcinomas, the cancer/testis(Cancer/Testis, CT) associated antigen gene. We aimed to analyze the expression level of PMR1 in gastric cancer tissues with poorly differentiatedã€moderately differentiatedã€well differentiated and its serum level in the preoperative and postoperative. Identify its value in the diagnosis and prognosis of gastric cancer and the meaning of the follow-up.54 gastric adenocarcinomas patients accepted surgical treatment in China-Japan Union of Jilin University in January 2013 to December 2015 as experimental group; 40 volunteers who have a health examination as control. All objects included after informed consent. The control collect blood samples only; experimental group collect preoperative and 7 days of postoperative blood samples.Meanwhile, collect patients with gastric cancer and normal gastric tissue during the surgery. RT-PCR detection of the gastric cancer tissues and normal gastric tissue PRM1- m RNA expression level; immunohistochemical staining analysis PRM1 protein expression levels in gastric cancer tissue and histological characteristics; Detect the PRM1 in peripheral blood in patients with gastric cancer and control by ELISA.(1)Poorly and moderately differentiated adenocarcinoma PRM1 expression levels higher than normal gastric tissue; while, well differentiated adenocarcinoma have no significance difference with normal gastric tissue.(2) Immunohistochemical staining detection PRM1 protein expression, the results showed that normal gastric tissue cell gap did not see positive staining; some of the well differentiated adenocarcinoma did not see positive staining, and others can see positive staining; in moderately differentiated adenocarcinoma, tan positive staining can be seen in the pulp; tawny dye can be seen in the pulp of poorly differentiated adenocarcinoma.(3)The Gastric cancer patients with peripheral blood PRM1 is significantly higher than that of healthy controls, patients with gastric cancer after tumor resection 7 days compared with preoperative serum levels of PRM1 decreased obviously. The serum level of PRM1 of poorly differentiation is lower than that of moderately differentiation and well differentiation, and difference between moderately differentiation and well differentiation is not obvious. Different clinical stages(phase I and phase II, phase III, IV) gastric cancer patients with preoperative serum PRM1 difference is not obvious.(1)PRM1- m RNA and its protein expression in gastric adenocarcinomas tissue is significantly higher than normal gastric tissue;(2) PRM1- m RNA and its protein expression level have certain correlation with the degree of differentiation of gastric adenocarcinomas.(3)PRM1 in serum of patients preoperative is significantly higher than control; postoperative day 7 significantly lower than the preoperative. Suggesting that PRM1 may be a serum marker as gastric cancer diagnosis, prognostic and follow-up. |
