Objective: The application of Shengjing Decoction in the treatment of idiopathic kidney essence deficiency type of infertility patients with oligospermia.By comparing the changes of sperm detection index before and after treatment,we observed the effectiveness of Shengjing decoction in the treatment of idiopathic kidney essence deficiency type of infertility patients with oligospermia,explored the correlation between the expression level of messenger ribonucleic acid(PRM1mRNA)of protamine 1 and oligospermia,explored its possible mechanism,and provided support for the popularization and application of Shengjing decoction in the treatment of infertility.Methods: A total of 36 idiopathic kidney essence deficiency type infertile patients with oligozoospermia who were treated at the Reproductive Maternal and Child Hospital of Chengdu University of Traditional Chinese Medicine from October 2018 to October 2019 were collected as treatment group.A total of 12 men with normal semen routine examination as control group.The treatment group was treated with Shengjing decoction Mianjian granules,which were provided by the Chinese Pharmacy of the Teaching Hospital of Chengdu University of Traditional Chinese Medicine for a total of 3 months.The efficacy of Shengjing Decoction in the treatment of idiopathic kidney essence deficiency type of oligospermia was evaluated by comparing the changes in sperm concentration,sperm forward movement rate(PR),and TCM syndrome scores in the treatment group before and after treatment.To investigate the possible correlation and mechanism between PRM1 mRNA and oligozoospermia by comparing the changes of sperm PRM1 mRNA expression level before and after treatment.The test data was statistically analyzed and processed using SPSS 20.0 software:during data analysis,the group of quantitative data that met the normal distribution was tested by t test,the comparison of the mean between groups was analyzed by single factor analysis of variance,and the non-parametric test that did not meet the normal distribution was used.The chi-square test was used for comparison of the rates,and the rank-sum test was used for variance inconsistencies.P <0.05 was statistically significant.Results:1.The mean sperm concentration before treatment in the treatment group was8.19±3.80×106/mL,and that in the control group was 78.12±36.20×106/mL.The sperm concentration before treatment in the treatment group was significantly lower than that in the control group,and the difference was statistically significant(P <0.05).The sperm PR before treatment in the treatment group was 24.79±8.38%,and that in the control group was 43.45±9.61%.The sperm PR before treatment in the treatment group was significantly lower than that in the control group,and the difference was statistically significant(P < 0.05).The mean expression level of PRM1 mRNA of sperm in the treatment group before treatment was 5.42±0.81×102copy/mL,and that in the control group was 18.36±2.55×102copy/mL.The expression level of PRM1 mRNA of sperm in the treatment group before treatment was significantly lower than that in the control group,and the difference was statistically significant(P< 0.05).2.The sperm concentration of the patients in the treatment group was8.19±3.80×106/mL before treatment,and 21.24±17.52×106/mL after treatment.The sperm concentration of the patients in the treatment group was significantly improved after treatment,and the difference was statistically significant(P < 0.05).In the treatment group,the sperm forward movement(PR)before treatment was24.79±8.38%,and the sperm forward movement(PR)after treatment was32.27±9.30%.The forward movement rate(PR)was significantly improved after treatment,and the difference was statistically significant(P < 0.05).3.The expression of PRM1 mRNA in the treatment group was5.42±0.81×102copy/mL before treatment,and 17.01±2.34×102copy/mL after treatment.The expression level of PRM1 mRNA in the sperm of patients in the treatment group was increased after treatment,and the difference was statistically significant(P < 0.05).4.The TCM syndrome score in the treatment group was 11.47±4.10 points beforetreatment,and 3.87±1.66 points after treatment.The TCM syndrome score was significantly reduced after treatment,and the difference was statistically significant(P< 0.05).5.According to the results of pregnancy condition and sperm concentration of subjects,the curative effect was determined.3 patients(10%)were cured,14 patients(46.67%)were showed to be effective,and 13 patients(43.33%)were effective.The total effective rate was 100% after treatment.According to the TCM syndrome score,the curative effect was judged.13 patients showed obvious effects,accounting for43.33%,14 effective patients,accounting for 46.67%,and 3 ineffective patients,accounting for 10.00%,and the total effective rate was 90% after treatment.Conclusion:1.Shengjing Decoction can increase sperm concentration and sperm forward motion rate(PR)in patients with idiopathic kidney essence deficiency type of oligospermia,and it has a definite effect in clinical treatment of idiopathic kidney essence deficiency type of oligospermia;2.The expression level of PRM1 mRNA is closely related to male reproductive function,and Shengjing Decoction can increase the expression level of PRM1 mRNA in patients with idiopathic kidney essence deficiency type of oligospermia;3.Shengjing Decoction can alleviate the symptoms of TCM in patients with idiopathic kidney essence deficiency type of oligospermia.
