The Influence Of Hongjindan On IL-2、IL-6 And JAK2/STAT3 Signaling Pathway Of Pelvic Inflammatory Disease Rats

Object:Through the experiments with rats, to observe the therapeutic effect of Hongjindan Enema on rats of pelvic inflammatory disease model,detected the level of IL-2,IL-6 in Pelvic inflammatory disease rats’ serum and the protein expression of IL-6,P-JAK 2,P-STAT3 in Rats’ uterine tissues,According this to explore the mechanism of Hongjindan’s treatment on pelvic inflammatory disease,to provide experimental basis and theoretical basis for clinical to treat the pelvic inflammatory disease with this kind of Chinese medicine.Method:After adaptive feeding a week,select 10 rats from all 60 rats 8 weeks of age as normal group(sham operated group),the remaining 50 successfully established as pelvic inflammation model by using phenol mucilage,then randomly divided into model group,Hongjindan in high、middle、low dose group(three groups)and Positive control group.After two courses of treatment(14days) with traditional Chinese medicine,took of the serum and uterine tissues of the rats,observed the morphology of rats’ uterus through eyes and light morphology.Detected the level of IL-2,IL-6 in rats’ serum by Elisa,detected the average optical density of IL-6 in rats’ uterine tissues by IHC and detected the protein expression of IL-6,P-JAK2,P-STAT3 in Rats’ uterine tissues by WB.Results:1 The influence of rats’ histomorphologyThrough the observation by eyes and light morphology can see that the pelvic adhesion degree of the model group was serious, the uterus congested and swelled obviously,adhere with the surrounding connective tissue and organ,can not be divided.The sham operation group and each drug administration group both had improved,especially Hongjindan in high dose group,Positive control group.2 The influence of the concentration of IL-2,IL-6 in rats’ serumThe level of IL-2 in model group was significantly lower than all groups else,had significant difference with them(P < 0.01).There was a certain degree of increase after treated by all groups treated with Chinese medicine,especially Hongjindan in high dose group,Positive control group,had no statistical significance with it(P>0.05).The level of IL-6 in model group was significantly higher than all groups else,had significant difference with them(P<0.01).There was a certain degree of decrease after treated by all groups treated with Chinese medicine,especially Hongjindan in high dose group,Positive control group and had no statistical significance with it(P>0.05).3 The influence of protein expression of IL-6,P-JAK2,P-STAT3 in Rats’ uterine tissuesThe average optical density of IL-6 in model group was significantly higher than all groups else,had significant difference with them(P<0.01),there was a certain degree of decrease after treated, especially Hongjindan in high dose group,Positive control group,had no statistical significance with sham operated group(P>0.05).The protein expression of IL-6,P-JAK2,P-STAT3 in model group was significantly higher than groups else,so had significant differences with them(P <0.05),compared with Hongjindan in high dose group had significant difference(P<0.01),there was a certain degree of decrease after treated,especially Hongjindan in high dose group and Positive control group, had no statistical significance with it(P>0.05).Conclusion:Hongjindan could obviously reduce the uterine swelling,congestion and the degree of adhesion with surrounding tissue of pelvic inflammatory disease model rats,thus promoted the absorption of uterine inflammation and recovered uterine morphology to treat pelvic inflammatory disease.the mechanism of Hongjindan’treatment on pelvic inflammatory disease was probably through increasing the level of IL-2,increasing the body’s immune capacity to achieve the treatment of pelvic inflammatory disease on one hand,on the other hand,inhibition of the expression of IL-6,decrease the protein expression of P-JAK2,P-STAT3 in Rats’ uterine tissues,and then achieved the treatment of pelvic inflammatory disease.
