Objective:Exploration of the WW domain E3 ubiquitin ligase WWP1 in breast cancer tissue expression, analyze the relationship between the expression and the clinicopathological factors of breast cancer,explore the relationship between ER, PR, p53 gene and the expression of HER-2, age, lymph node metastasis, histological grade and tumor size WWP1 gene and breast cancer patients, in order to provide evidence for judging the clinical treatment and prognosis of breast cancer.Materials and methods:from 2004 March-June 2007 accepted breast cancer resection of 129 cases of women patients with breast cancer paraffin embedded specimens, the application of tissue microarray technology construction of breast cancer by tissue microarray and he staining to determine the quality of tissue microarray, immunohistochemistry staining of WWP1 observation was. Select another breast benign lesion resection in 19 cases of female patients with breast tissue paraffin embedded specimens were used as controls. The breast cancer patients with age, tumor size, axillary lymph node metastasis, pathological grade and estrogen receptor, progesterone receptor and EGF receptor 2 (HER-2) gene expression and the WWP1 expression for the correlation analysis.Results:HE staining showed that the two groups of tissue microarray quality is good, can represent the related disease organization pathological morphology. Immunohistochemical staining showed that WWP1 expression was positive in 92 cases (70.5%) in the breast cancer tissues, and the expression of WWP1 was not found in the benign breast tissues. The expression of estrogen receptor, progesterone receptor and WWP1 positive expression rate differences were not statistically significant (P> 0.05), tumor size and the WWP1 positive expression rate was positively correlated (P< 0.05); axillary lymph node metastasis and HER2 gene expression positive of WWP1 positive expression rate was significantly higher in the negative expression of patients (P< 0.05); Conclusion:WWP1 is overexpressed in breast cancer tissues and is negatively correlated with histological grade、ER、PR expression and Survival prognosis. WWP1 is overexpressed in breast cancer tissues and is positively correlated with lymph node metastasis and Her-2 expression. It has the potential to be a new prognostic marker and therapeutic target of the breast cancer.
