Objective: To investigate the effect of calcium gluconate on the expression amount of pain factors including P2X3, CGRP, COX2 and IK1 in the rat spinal cord induced by Latrodectus tredecimguttatus venom. This study have confirmed calcium gluconate treatment effect of pain. Methods: 30 male SD rats were randomly divided into pain group(15) and calcium treatment group(15), the treatment group rats were in advance injected 10% calcium gluconate 2mL in tail vein, pain group rats injected saline 2 mL, then the two groups of rats injected 5% formalin solution for 150 m L and 4 kinds of concentrations of Latrodectus tredecimguttatus venom on plantar. Then rats were killed after 24 h and got the spinal cord L4~5, Using immunohistochemical method, Western blotting method, real-time fluorescence quantitative PCR technology analyzed expression of CGRP, P2X3, COX2 and IK1 in different respectively. Results: In the venom dose of 100 m L/kg and 300 m L/kg, Compared with the pain group, the expression of CGRP and P2X3 in the treatment group decreased significantly(P<0.05) in previous tow methods. Real-time f-luorescent quantitative PCR resugts showed that P2X3, CGRP and COX2 mRNA e-xpression in the treatment group were higher than pain group, difference has no st atistical significance(P>0.05), the difference of expression of CGRP, P2X3, COX2 and IK1 in each pain groups has no statistical significance(P>0.05). the difference of expression of the pain factor in formalin pain and treatment group was statistically significant(P<0.05). Conclusion: The possible mechanisms of pain induced by Xinjiang Latrodectus tredecimguttatus venom may be related to the level of expression of pain factors in rat spinal cord including CGRP, P2X3, COX2 and I-K1, calcium gluconate cougd significantly inhibit the relative expression of pain factors induced by high concentration toxin and the transmission of pain.
