Objective: We detected the expression of TBK1 and NF-κB rel A in 80 cases of hepatocellular carcinoma tissues, 80 cases of corresponding noncancerous tissues and 30 cases of normal liver tissue. Combining with clinical pathological data,we investigated the possible mechanism of TBK1 and NF-κB relA in HCC occurrence and development, as well as the correlation of their expression in HCC.Methods:(1) We collected 80 cases of HCC resection and 80 cases of adjacent cancer tissues(more than 2 cm from cancer tissue),from the Department of pathology of Zhengzhou University Affiliated Tumor Hospital in Henan Province during 2009-2015, and another 30 cases of patients with hepatic hemangioma by surgical resection as the normal control group. Then immuno histo chemical method(SP method) was used to detect the expression of TBK1 and NF-κB relA in these tissues.(2) We collected 32 cases of HCC resected fresh tissues and 32 cases of fresh tumor adjacent tissues, which were 2 cm from surgical resection margin, from the Department of hepatobiliary surgery of Zhengzhou University Affiliated Tumor Hospital in Henan Province during 2011-2015, and also collected 10 cases of fresh liver tissues resected from hemangioma treated by surgical resection asthe normal control group. Real-time fluorescence quantitative gene amplification method(RT-PCR) was used to detect the expression of TBK1 and NF-κB relA mRNA.(3) We analyzed the correlation between TBK1 and NF-κB relA, and their relationship with the clinical pathological data.Results:(1) The positive expression rate of TBK1 protein in HCC tissues, paracancerous tissues, normal tissues were 65.0%(52 / 80), 16.3%(13/80) and 10%(3 / 30) respectively. The expression in the HCC tissues was significantly higher than in adjacent tissues(P < 0.05) and normal tissues(P < 0.01), and the difference was statistically significant. TBK1 protein expression in hepatocellular carcinoma(HCC) was related with the hepatitis B surface antigen(P < 0.05), the expression of TBK1 positive is high in hepatitis B surface antigen positive group. But it had no significant correlation with gender, age, AFP, tumor diameter, portal vein tumor thrombus or tumor differentiation(P > 0.05);(2) The positive expression rate of NF-κB relA protein in HCC tissues, paracancerous tissues, normal tissues were 67.5%(54/80) 20.0%(16/80), and 6.7%(2 / 30) respectively.The expression in the HCC tissues was significantly higher than in adjacent tissues(P < 0.05) and normal tissues(P < 0.01), and the difference was statistically significant.NF-κB relA protein expression in hepatocellular carcinoma(HCC) was related with tumor differentiation(P < 0.05), The expression of positive NF-kappa B rel A protein is high in the poorly differentiation group. But it had no significant correlation with gender, age, AFP, tumor diameter, hepatitis B surface antigen or portal vein tumor thrombus(P > 0.05).(3) TBK1 expression and NF-κB relA expression in HCC tissues were positively correlated(r=0.274, P =0.014).(4) The TBK1 mRNA in HCC tissues was significantly higher than that in carcinoma adjacent tissues and normal tissues(P < 0.05). And its expression had relationship with surface positive antigen hepatitis B(P < 0.05), The expression of positive TBK1 is high in hepatitis B surface antigen positive group. But had no significant correlation with gender, age, AFP, tumor diameter of portal vein tumor thrombus or tumor differentiation.(P > 0.05)(5) The NF-κB relA mRNA in HCC tissues was significantly higher than that in carcinoma adjacent tissues and normal liver tissues(P < 0.05).NF-κB relA mRNA in HCC was related with the tumor differentiation(P < 0.05), The expression of NF-kappaB rel A mRNA is high in the poorly differentiated group. But it had no significant correlated with gender, age, AFP, tumor diameter, hepatitis B surface antigen or portal vein tumor thrombus(P > 0.05). Conclusion: 1.TBK1 and NF-κB relA are involved in the progression of HCC.The high expression of TBK1 in HCC tissueis related with the infection of hepatitis B virus. And NF-κB relA high expression is correlated with tumor differentiation. TBK1 and NF-κB relA are positively correlated in expression.2.TBK1 and NF-κB relA have the potentiality to be the index for the diagnosis of hepatocellular carcinoma, and it could provide basis for HCC related forensic identification.
