Purine metabolic disorders will lead to increasing of the production of uric acid, this role or accompany uric acid excretion dysfunction in kidney will raise the level of uric acid in the blood, then caused hyperuricemia. Most mammals have uric acid enzyme, uric acid can be converted to allantoin, and then easily through out with the urine. Because of the lack of uric acid enzyme, and the end product of purine metabolism is still the uric acid in human. The high level of blood uric acid causes hyperuricemia, and lead to urate crystal deposition in the joints and kidneys. It is reported that taurine (2-amino-ethanesulfonic acid, beta taurine) has a wide range of biological functions and participates in several biologic processes in the kidney. Taurine is an endogenous amino acid of the sulfur-containing, it is necessary study the effect of taurine on hyperuricemia. Our laboratory pilot study found that the effect of taurine on hyperuricemia nephropathy rats is reducing kidney damage and promote the uric acid excretion. But the mechanism is unclear. This study aimed to investigate the effects of taurine on uric acid excretion and explore their underlying mechanisms in hyperuricemia nephropathy rats, which were induced by adenine (200 mg/kg/d) and ethambutol hydrochloride (250 mg/kg/d) by using stomach tube to SD rats, and adding 1%,2% taurine in the drinking water.Experimental rats were randomly divided into 6 groups:normal control group,2% taurine control group, model group, drug group,1%and 2% taurine group. The drinking water of the taurine group and drug group orally administered taurine and drug. The drinking water of normal control group and model group rats is normal. Then, adenine and ethambutol hydrochloride were intragastrically given to rats for ten days except control group and 2% taurine control group. And detect the pathological anatomy changes of kidney, renal biochemical index, NO level, oxidation resistance and urate metabolism transporters URAT1, UAT and MRP4 gene mRNA and protein relative expression level.The results of the test showed that①taurine can significantly reduce the renal indexl, renal diseade points, the levels of serum uric acid (UA), creatinine (Cr) and urinary protein excretion, increase the excretion of urea nitrogen (BUN) of the hyperuricemia nephropathy rats (P< 0.05), and close to the normal level (P> 0.05).②taurine can significantly make hyperuricemia nephropathy rats nitric oxide enzyme(NOS), and close to the control level (P > 0.05). Taurine have no significant impact on the serum nitric oxide (NO), and significantly increase the T-AOC and SOD, close to the control level (P> 0.05), on the contrary, raurine reduced the content of MDA (P< 0.05), close to the control level (P> 0.05).③taurine can significantly increase hyperuricemia nephropathy rats kidney UAT, MRP4 gene and protein expression levels (P< 0.05) and reduce URAT1 genes, URAT1 protein expression levels tend to be normal (P> 0.05).In summary, taurine can effectively prevent gouty nephropathy model in rats, the action on taurine may be relieve renal function injury and regulate the excretion of uric acid related gene.
