Objective:By behavior experiment,HE staining and CaMKⅡ expression inhipp-ocampal neurons were studied to investigate therapeutic effects of Jiann-aoyizhi Capsule,which based on improving microcirculation and decrease brain anoxia and brain edema.Methods:Option SD rats(8 weeks old) were randomly divided into five group-s,namely group A(sham-operated group),group B(model group),group C(traditional chinese medicines group),group D(piracetam group),group E(control group).Single brain samples collected by timing divided into subgroups s uch as the A1-A5;Each group electronic brain injury instrument(e CCI) were TBI modeling.A group(do sphenotresia,see the whole dura,do not fight the dura);group B(routine feeding after craniocerebral injury);group C(give traditional chinese medicines orally after craniocerebral injury,Jiannao Y-izhi capsule is administered 3 times/day,specs:0.75g/ml suspension);D gr-oup(give piracetam orally after craniocerebral injury,piracetam tabletsa-dministered 3 times/day,specs:0.09g/ml suspension);E group(give only ro-utine feeding).Using Morris water maze observe and record the impact of the experiment before modeling and model rats were given appropriate inte-rvention after the model of learning and memory and other acts 1st,3rd,5-th day,7th day,14 th day of school,and at the specified node respectively1 st,3rd,5th day,7th day,14 th day in the corresponding subgroup collect br-ain tissue samples.The HE staining result were observed by the microscop-e,immunohistochemistry study of CaMKⅡ.Results:The experiment was a success modeling,according to NSS score,all brain function in rats were badly damaged.A total of 48 rats were lethal(mortality rate 27.0%).All rats before craniocerebral injury brain functi-on are complete,and in the first day learning and memory ability is poor after craniocerebral injury.1.Morris water maze results:The model group at each time point escape latency was significantly longer than theother groups,and crossing platform times were significantly less than theother groups.The difference was statistically sicgnifiant(P<0.05);there was no statistically sicgnifiant that we are compared all of the B,C,D groups da-tebases with each other;the 5th days,group B and group C were statistica-lly sicgnifiant(P<0.05),and group C was better than group B;no significa-nt difference(p>0.05)in group B and group D,no significant difference(p>0.05)in group C and D groups.In the 7th day,group B and group C,D grou-ps were different,but no difference in group C and D;the 14 th days B,C,D twenty-two three groups were different.Wherein latencies:B group>A group>D group>C group,and crossing platform times:B group<A group<D group<C gro-up,with statistical significance;2.HE staining result:in 1st-5th day a large number of necrosis and inflammatory cell infiltration were seen aro-und the lesion,C group,D group were compared with other groups of newbor-n neurons and microvascular composition;0n the 7th-14 th day,the edema of neurons was found to decrease than before.However,there are still a large number of inflammatory cell infiltration and proliferation of glial cells,group C which neurons and the number of new blood vessels increased is be-tter than group B.Model group than the other groups seriously;3.The CaMK-Ⅱ average optical density comparison:A and E groups was not statistical-ly significant,B,C,D brain tissue in rats within 1st-3rd day detected high expression of CaMKⅡ(p>0.05),no significant differences.The 5th day,again detected CaMKⅡpositive rate of cells in each group rat brain tissu-e begins todrop,group B and group C(P<0.05)differences in group C than group B;No difference between group B and group D(p>0.05),group C and D w ere no differences(p>0.05).In the 7th to 14 th day,group B CaMKⅡ detected expression level is still higher than other groups,B,C,D pairwise compari-son,(P<0.05) there are different,where are in average optical density:B group>D group>C group>A group,and there are significant differences(P<0.05).Conclusion:1.By behavior experiment,Jiannaoyizhi Capsule was proved that it has therapeutic mechanism that increase the learning and memoryizing ability;2.Jiannaoyizhi capsule can promote hippocampal neuron regeneratio-n and stable environment to improve the nerve cell survival rate,the inc-rease in the number of neurons,and brain dysfunction is improved,so as to improve the learning and memory impairment;3.Jiannaoyizhi Capsule can inhibit CaMKⅡ high expression and prevent a decline in the activity of CaMKⅡ,or induced the enzyme activity recovery,enhance synaptic transmiss-ion,reduce or inhibit the neuronal damage,protect of brain function and promote the recovery of learning and memory impairment after brain tissue injury.
