Objective: This experiment was aim to reveal whether the Jiannaoyizhi capsule can get the injured brain tissue microcirculation be better.we compared the different groups by observing the general state of the craniocerebral injury rat at the different time points and analyzing the conent of the neuropeptide Y (NPY) and calcitonin gene-related peptide (CGRP) which exiting in the plasma of the rats in the corresponding group.Methods: The SD rats were given the corresponding number and divided into 4 groups randomly.The were the A group (JiaShouShu group),the B (model group), the C (Naofukang treatment group), the D (Jiannaoyizhi capsule treatment group), and each group were divided into 4 subgroups randomly by taking blood at the different time points. In addition to the A group , the others were made moulds based on the Feeney method[1]. The rats in A group were only exposed the dural mater and not to strike,then sewing up the scalp,feeding rountine, not giving the other therapeutic treatment. the B group was made moulds,and the feeding was same to the A group. The C group was fed with the Naofukang,the drug was thawed into warm water and becoming the suspension with the denisity in the 0.09g/ml,they were given the rats with the dose at 1.2g/g/time by gavaging,3 times/day.The D group was same to it,the Jiannaoyizhi capsule was thawed into warm water and becoming the suspension with the denisity in the 0.75g/ml,they were given the rats with the dose at 1.5g/g/time by gavaging.To observe the corresponding objective response in rats situation(mental status, food intake,behavioral activity)at the modeling before and after treatment 1.3.5.7.10 days.what, s more,according to the specified time points 3.5.7.10 days,the rats were sacrificed in the corresponding section,and taking blood from the right atrial, then the content of the NPY and CGRP would be measured(by the method of RIA)and analyzed.Results: Successful modeling of the experiments, according to the rats general state,it belongs to the middle-severe traumatic brain injury.There were 44 rats died (mortality 31.4%)in the experiment. Observing the rats in each group, they were well before making model and on the first day after modeling, they are apathetic, unresponsive, the B, C, D were obviously;At the third day,A, C,D group showed improvement over the previous,B group had no obvious change; At the fifth day,every group has improved performance, but the B group was poor;At the seventh and tenth day,the A and D group rats have the obviously progress than the previous reaction, B and C group had varying degrees of progress. Analysis of data showed that: D group and C group index changes in plasma early after injury (3 days) no significant difference (P> 0.05), compared with B group,the NPY and CGRP values lower (P <0.05), they were different; after injury (5-10 days),D group is lower than the C group of the NPY, the CGRP was higher than C group (P <0.05),they were different,compared with group B ,the NPY was significantly lower and the CGRP value at a high level (P<0.05),there were significant differences.Conclusion:1. Jiannaoyizhi capsule can adjust the level of NPY and CGRP on craniocerebral injury rats and reach the aim of getting injured brain microcirculation better.2.The possible mechanism: (1)To adjust the reaction of the sympathetic, inhibit the excessive release of the NPY and regulate the change of the CGRP, to get the cerebral microcirculation better.(2)To promote the blood brain barrier repaired quickly and inhibit the Ca2+ internal flow excessively, in order to reduce the cells damage because of the calcium overload.
