| Objective: Preliminary exploration of xinjiang syphilis patients with treponema pallidum(TP), the characteristics of the genotypes for high incidence of syphilis in Xinjiang provide theoretical basis for molecular epidemiology research.Methods: Collect blood specimens from 95 patients with syphilis infection, treponema pallidum genomic DNA extracted with the use of microbial genome extraction kit, ordinary PCR amplification TP specificity polA genes, screening of positive specimens, from positive samples containing TP DNA, amplification for TP genotyping of arp, TPR, tp0548 genes, after the success of the expansion, according to the principle of the genotyping, TP genotyping, ultimately determine the TP genotypes in Xinjiang.Determined using ordinary and nested PCR amplification Tp blood samples.95 patients with early syphilis were extracted with commercial kits of TP DNA in whole blood specimens, using ordinary and nested pol A polymerase chain reaction(PCR) amplification, after ordinary PCR screening polA 25 cases, 25 cases verified by further nested PCR positive for 10 cases and increase positive specimen.Use the 35 cases of the new TP in whole blood specimens arp, TPR, tp0548 gene, analysis of TP genotyping. Results: 95 syphilis blood specimen, TP and 35 cases specific DNA positive rate of 36.8%, 35 cases of positive specimens, the arp, TPR, tp0548 gene amplification, 10 cases were successful, 10 cases of specimens using TP genetic classification, completely can complete classification, including 8 patients with han TP genotyping for 14d/f, 1 case of kazak patients TP genotype for 14d/f, 1 case of Uighur patients TP genotypes of 15d/f.Conclusion: Xinjiang syphilis patients with TP genotypes are mainly for 14d/f treponema pallidum gene subtype, successful classification in patients with different nationalities can get 8 patients with han genotyping for 14d/f, 1 case of patients for 14d/f, 1 case of Uighur patients for 15 d/f. |
