Objective To investigate the effect of interactions between genes and gene,environmental factors on hyperuricemia,we used different statistical methods like Logistic regression,MDR,and cross-sectional analysis.This study will establish early foundation in Pathogenesis of hyperuricemia,and also Provide a theoretical basis for the study of genetics of hyperuricemia among different ethnic groups in Ningxia Hui Autonomous Region.So as to adopt a more scientific and reasonable Prevention and treatment method for hyperuricemia groups.Methods(1)Subjects: From May to October 2014,the People who underwent health examination in a medical examination center in Yinchuan,Ningxia Hui Autonomous Region were selected.According to the inclusion and exclusion criteria,362 unrelated hyperuricemia Patients were eventually classified as case groups.Based on age(disParity ≤ 5 years old),gender and nationality,a 1:1 matched case-control study was used.At the same time,362 healthy People with similar background data were selected as the control group.General demographic characteristics: in the case group,229(63.3%)were males,133(36.7%)were females,194(53.6%)were Huis,168(46.4%)were Hans,and the range of age was 29 to 80 years.The average age was 56.71±9.39.The age range of control group was 28 to 81 years,and the average age was 56.68±9.46 years.(2)Research methods: Trained qualified investigators collected demographic characteristics,medication status,and other relevant data through questionnaires,Physical measurements,and laboratory biochemical indicators.Physical measurement indicators include height,blood Pressure,body weight and so on.Laboratory biochemical detection and DNA extraction through the collection of venous blood.The laboratory detection indexes include(SUA,Crea,TC,TG,BUN,HDL and LDL),etc.Used the Sequenom Mass ARRAY iPLEX GOLD technique to detect single nucleotide Polymorphisms in the rs6811287 and rs7663032 of SLC2A9,the SLC22A12 rs559946 and LRRC16 A rs742132.Test whether the frequency distribution of genotypes has Population genetic representation through the Hardy-Weinberg equilibrium law test.Comparison of the mean values between different groups of biochemical indexes by Paired t-test.Chi square test was used to compare the distribution of genotypes and alleles in different groups.Used the Multivariate Logistic Regression to explore the influencing factors of Hyperuricemia.The interaction between rs6811287 loci and rs7663032 loci of SLC2A9 gene,rs559946 loci of SLC22A12 gene and LRRC16A’s rs742132 loci was analyzed by MDR software,cross analysis and conditional Logistic.Results:1.Comparison of biochemical indexes between two groups Crea,TC,TG,SBPand BMI were higher in the case group than in the control group(P<0.05).There was no significant difference inBUN,FBG,HDL,LDL and DBP between the two groups(P>0.05).2.ComParison of Genotype Frequency between two groups The genotype frequencies of SLC22A12 gene rs559946,SLC2A9 gene rs6811287 and rs7663032,and LRRC16 A gene rs742132 were not significantly different between the two groups(P>0.05).3.In the case group,The distribution of genotypes and alleles of rs742132 loci in LRRC16 A was statistically significant between males and females(P<0.05).There was no significant difference in genotype distribution and allele distribution among all SNP loci of SLC22A12,SLC2A9 and LRRC16 A of different nationalities and sexes(P>0.05).4.Multivariate Logistic Regression Analysis Crea,TC and BMI as risk factors for hyperuricemia by Logistic regression analysis(P<0.05).5.Interaction analysis Based on the multifactor dimensionality reduction,the results showed 3 fators(SLC22A12-rs559946,LRRC16A-rs742132 and Triglyceride)model be the best of gene-environment analysis because of higher cross-validation consistency and accuracy of test sample.The accuracy of test sample of best model is 0.5912,P =0.1071,there were no statistically significant difference in the model.It is suggested that there is no interaction between rs559946,rs742132 and triglyceride.Based on the logistic regression analysis revealed that SLC2A9 gene rs6811287 site increased the risk of hyperuricemia,and there was a multiplicative interaction between SLC22A12-rs559946 with LRRC16A-rs742132.By crossover analysis,after statistical testing of the model,the result showed that LRRC16A-rs742132 with triglyceride was significant interacted in addition model(U=2.146,P<0.05).There were significant Positive interaction between SLC22A12-rs559946 with LRRC16A-rs742132 in multipcative model(Orint=1.287,P<0.05).Conclusions: 1.In the case group the frequency and allele of the rs742132 loci in the LRRC16 A gene was statistically significant between male and female.2.Crea,TC,and BMI are risk factors for Hyperuricemia.3.Interaction analysis showed that SLC2A9 gene rs6811287 site is a risk factor of hyperuricemia,There were Positive interaction between SLC22A12-rs559946 with LRRC16A-rs742132 in multi Pcative model,and LRRC16A-rs742132 with TG in addition model. |
