Actinidia eriantha Benth. belongs to the family of Actinidia and the genius of Actinidia. It mainly attributes in provinces such as Zhejiang and Guangxi in China. Its roots can be pick up all the year round, while its leaves in summer and fall. According to the record of Chinese Pharmacopeia, it has various medical effects including curing the cancer of stomach, nose, and breast. Current references focus on its chemical components and biological activity, and the former mainly include triterpenes and polysacchrides while the latter mainly include anti-cancer and immune modulation.This study systematically carried experiments on the chemical components of Actinidia eriantha Benth. and its effects on inhibiting the proliferation of human tumor cell lines. Samples (mainly its root and stem dried slices) were collected from Suichang in Zhejiang province in October,2008, and identified by associate professor CHEN Xilin of Zhejiang Chinese Medicine University.1 kilogram sample was smash and extracted by the 80% methanol solution with the assistance of microwave. The methanol was recycled. And then chloroform, ethyl acetate and 1-butanol saturated by water were sequencely used to extract it. After that, all these organic solvent were recycled and the according extracts (designated as Chlo, EtOAc, n-BuOH, respectively, and the total extract by methanol as MeOH) were obtained and froze-dry. The weight percentages of the three extracts were 0.13%,0.07% and 0.58%, respectively.We examined the in vitro anti-cancer activity of the above extracts through MTT, determining the most effective extract and the sensitive cancer cell lines. The cell lines used are liver cancer SMMC-7721, lung cancer A549, breast cancerMDA-MB-453, colon cancer HT-29 and esophagus cancer TE-1. The normal liver cell line L-02 as the positive control, and adriamycin (abbreviated as ADM) as the positive drug control. The result showed that Chlo extract is most effective in inhibiting cancer cell growth, and then is the EtOAc and n-BuOH extract. Within the set drug dose, for the Chlo extract the sensitivity sequence of cancer cell line is SMMC-7721>TE-1>HT-29>MDA-MB-453>A549; and for the EtOAc extract is HT-29>SMMC-7721>A549>MDA-MB-453>TE-1; and for the n-BuOH extract is MDA-MB-453> HT-29> TE-1> SMMC-7721>A549; and for the total extract is SMMC-7721> HT-29> MDA-MB-453> A549>TE-1. The IC50 of Chlo extract for SMMC-7721, HT-29, TE-1 and MDA-MB-453 post 72h were 221.82μg-mL-1,303.39μg·mL-1, 224.39μg·mL-1 and 315.50μg·mL-1, respectively. The IC50of EtOAc extract for SMMC-7721 and HT-29 post 72h were 563.64μg·mL-1 and 483.06μg·mL-1, respectively.In the study of Chlo extract which has the most effective anti-cancer activity,15 samples were collected through silicon thin-layer chromatography and silicon column chromatography. According to the color reaction, the major substance of Chlo extract belonged to alkaloids. Then the No.2 sample was examined through analytical HPLC, and the result showed that peaks appeared when the concentration of acetonitrile was 85% and the retention time was at the duration of 8.5-10min. But they were not isolated thoroughly. The color reaction showed that the major substance of EtOAc and n-BuOH belong to flavonoids All the work above made some preparation for further isolating the possibly anti-cancer single chemical compound.
