Actinidia eriantha Benth, derived from kiwi plants, is the root of eriantha Benth. The dried roots are widely distributed in the provinces of Zhejiang, Fujian, Jiangxi, Hunan, Gua-ngxi and Guangdong. According to Chinese Dictionary and the National Chinese herbal me-dicine records It was used to clear heatness and dampness, alleviate blood swelling, detoxi-fication and, traditional for the treatment of gastric cancer, breast cancer, esophageal cancer, and uterine body cancer.The tumor is one of the important diseases that threaten human health, clinical surgery, chemotherapy, radiation therapy are the primary means for the treatment of tumors,which Would cause kinds of shortcomings due to the side effects of these treatments for example damage to the human body, the majority of patients were difficult to tolerate, looking for drugs and methods that would be effective for cancer treatment is an important issue in the current anti-cancer research field. Chinese medicine has a great advantage in the treatment of cancer, with surgery in the prospects of alleviating the toxicity of radiotherapy、chemo-therapy, improving the treatment efficiency and prolong survival time,Chinese medicine has shown broad application.This article determined the root of Actinidia eriantha Benth, hair flower, total flavonoids, total phenols and total triterpene acids ultrasonic extraction process and content by ultraviolet spectrophotometry.The experimental results of total flavonoids optimum extraction technology of extraction time30min, extracted3times, solid-liquid ratio is1:30,80%ethanol; optimum extraction of total phenols ultrasonic extraction time30min, extraction times2, solid-liquid ratio is1:30, the ethanol concentration of60%; total triterpene acid best ultrasonic extraction process for the extraction time30min, extracted3times, solid-liquid ratio is1:20; crude drug, total flavonoids, total phenol, total triterpene acid content were1.092%,0.991%,1.19%.four human tumor cell models, SGC-7901, MGC-803, RKO and HepG2were used screen cytotoxicity with Actinidia eriantha Benth ethanol extract and four polar extract fractions by MTT method. The results indicated that EE-AER exhibited the strongest cytotoxicity against human tumor cells than other extracts and SGC-7901cells showed the most sensitive to EE-AER; that100μg/mL of ethyl acetate extract(EE-AER) most pronounced inhibition of SGC-7901cells, the inhibition rate of92%, respectively (P<0.01). Especially SGC-7901cells were shown to be the most sensitive to EE-AER with time-and dose-dependent manner. EE-AER could inhibited the proliferation of SGC-7901cells significantly with the IC50value of57,41,35μg/ml in24,48,72h, respectively (P<0.01); Furthermore, apoptosis in SGC-7901cells induced by EE-AER was detected by flow cytometry and DNA fragment assay,The results showes that it can induce apoptosis in SGC-7901cells, DNA ladder was observed by agarose gel electrophoresis; flow cytometry results found that different concentrations (40,60,80μg/mL) ethyl acetate extract can induce apoptosis in SGC-7901cells, and its role is enhanced with the increase in the concentration of administration; RT-PCR and Western Blot detection of different concentrations (40,60,80μg/mL) of ethyl acetate extract intervention SGC-7901cells, the cells of Bax, Bcl-2, Casepase-3expression also will be changed;addition, in Actinidia eriantha Benth the total flavonoids, total phenolics and total triterpene acids were determined by ultrasonic extraction process wit ultraviolet spectrophotometry. Total flavonoids, total phenol, total triterpene acid content in the roots were1.092%,0.991%and1.19%, respectively.In summary our preliminary experimental results showed that the root of Actinidia eriantha Benth exhibited significantly inhibitory activity against gastric cancer cells SGC-7901, and its anti-tumor substance basis and mechanism of action needs further study to provide strong scientific basis for the development and utilization in the anti-tumor research field.
