Actinidia polygama,a plant of the genus Actinidia belongs to the family Actinidiaceae,is mainly distributed in northeast China,north Chian,North Korea and Far East of Russia.Modern pharmacological studies on A.polygama indicate that it has anti-rheumatism effect,commonly used in the treatment of low back pain,stroke hemiplegia and other diseases.At present,the chemical composition study of A.polygama focus on the fruits,but rhizome related research little.In this study,we describe the analysis of chemical constituents from the rhizome of A.polygama,and their anti-inflammatory activities were evaluated,which enriched pharmacodynamic material basis of A.polygama and provided a scientific basis for making good use of this plant.In this study,ten compounds were obtained by means of various separation and purification technology such as HPLC,silica gel column chromatography,preparative thin layer chromatography,Sephadex LH-20 column chromatography and recrystallization from petroleum ether and the ethyl acetate fractions of rhizome of A.polygama,and they were identified as 3?-O-acetylursolic acid(1),a-linolenic acid(2),stigmast-4-en-3-one(3),lauric acid(4),?-sitosterol(5),palmitic acid(6),hexadecan-l-ol(7),woodwardic acid(8),daucosterol(9),and(-)-epicatechin(10)on the basis of spectroscopic(1H-NMR,13C-NMR,MS,ect)and physicochemical methods.Among them,compounds 1,2,3,4,6,7,8,9,10 were isolated from the plant for the first time.The isolated compounds were evaluated for their inhibitory effects on TNF-a-induced NF-?B activation to estimate anti-inflammatory activity in vitro,and cell viability was measured by MTT assay.As a result,none compounds exhibited significant NF-?B inhibitory activities.Compounds 1 and 8 had significant cytotoxicity to HeLa cells at 100?M,and other compounds did not show obvious cytotoxicity.
