Tibetan Medicinal Herb "Zhama" from root or stem bark of various plants in the Leguminosae sp. Caragana Fabr. have the function of clearing heat from muscles and meridians.Sichuan province is the second largest Tibetan region in China. According to the survey, "Zhama" of Sichuan Tibetan region are mainly from Caragana bicolor Kom.and Caragana erinacea Kom. At present, there is no quality control standards and the pharmacognosy studies also rarely been reported.To cooperate with establishing the "standard of Tibetan medicinal material in Sichuan Province" 2014 edition,this research systematically studied on the measures of quality control of Caraganae Bicorloris Radix and Caraganae Erinacea Radix, including the source, characters, identification (microscopic identification、TLC), checking (waters、total ash、acid insoluble ash).determination of extractives.In the meantime, Safety evaluation, anti-inflammatory and analgesic effect of Caraganae Bicorloris Radix.and Caraganae Erinacea Radix has been researched.And then on this basis, combined with the technical requirements of medicinal materials quality standard,we established the draft of quality standard of "Zhama". Some of the contents have been included in the "standard of Tibetan medicinal material in Sichuan Province"2014 edition.Main contents and results are as follows:1.This is the first detailed study on the morphological organization of Caragana bicolor Kom. and Caragana erinacea Kom.The height of plant,compound type of leaves, the amount of leaflets,the way of peduncle insertion,the color and pattern of the flower,and the other aspects of fruit can be identified from the Original plant.Medicinal material characters can be identified mainly from the size, color and texture of the surface, texture, fracture surface etc. Microscopic identification can be identified mainly from transverse section and powder.The transverse section can be identified mainly from the cells in cork layer, the proportion of phloem and xylem, crystal sheath fibers of phloem, the arrangement mode of xylem vessel and fibers etc.The powder can be identified mainly from the crystal sheath fibers, conduit and the square crystals of calcium oxalate.2.The TLC methods of "Zhama" has been established,which used control medicinal materials and β-sitosterols as their reference substances.The good separation of "Zhama" was taken by using the petroleum ether(60-9O℃)-ethyl acetate--formic acid (5:1:0.06) as developing solvent. The Rf values were moderate and the chromatographic spots were clear.The TLC tests of six batches Caraganae Bicorloris Radix and ten batches Craganae Erinacea Radix from different areas showed that the methods were adaptable and reproducible in identification,3.According to the "Chinese Pharmacopoeia" 2010 edition, we measured the impurity,moisture, total ash and acid insoluble ash of sixteen batches medicinal materials from different areas.According to the measured results, connected with practical production, the limit was developed as the follows:The impurity,water, total ash and acid insoluble ash could not be over2.5%,9.0%,5.0% and 2.0%, respectively.4.Comparison of different extraction methods and solvent, the methods that hot dipping method with 75% ethanol of extract determination for "Zhama" was established.According to the measured results of six batches Caraganae Bicorloris Radix and ten batches Caraganae Erinacea Radix medicinal materials from different areas,connected with Production of practical,the limit was developed as not less than 8.0%5. The chemical components of Caraganae Bicorloris Radix and Caraganae Erinacea Radix were preliminary tested by the combined methods of tube reaction, thin-layer chromatography with circular filter paper and thin-layer chromatography with silica gel g thin layer plate. According to the experimental phenomena, it was preliminary infered the Caraganae Bicorloris Radix contained Amino acids, peptides, proteins, polysaccharides, glycosides, saponins, organic acids, phenols, tannins, steroids, triterpenoid saponins,sterols,grease. The Caraganae Erinacea Radix contained amino acids, peptides, proteins, polysaccharides, saponins, glycosides, organic acids, flavonoids, phenols, tannins, steroids, triterpenoid saponins, sterols, grease.The research laid the foundation for follow-up study.6. This was the first study on safety evaluation, anti-inflammatory and analgesic effect of Caraganae Bicorloris Radix.and Caraganae Erinacea Radix, provided references for tibetan medicine clinical edication.6.1 The experimental results of safety evaluation showed that the maximum tolerance dosage of Caraganae Bicorloris Radix by gavage was more than 166.8g/kg, which was about 122 times of adult daily dosage. Caraganae Erinacea Radix was more than 150.0g/kg, which was about 109 times of adult daily dosage.6.2 According to studying anti-inflammatory action of Caraganae Bicorloris Radix and Caraganae Erinacea Radix with the model of mice auriculate tumefaction caused by xylene, we got the result, which showed the mouse auricle tumefaction degree of dexamethason group, Caraganae Bicorloris Radix high dose group and middle dose group had highly significant difference compared with model control group (**P<0.01); Caraganae Erinacea Radix low-dose group had highly significant difference compared with model control group (*P<0.02).6.3 According to studying analgesic effect of Caraganae Bicorloris Radix and Caraganae Erinacea Radix with the model of mice body torsion reaction caused by intraperitoneal injection glacial acetic acid, we got the result, which showed the incubation period of aspirin group and Caraganae Erinacea Radix high dose group had highly significant difference compared with model control group (**P<0.01).The writhing response frequencies of aspirin group,Caraganae Bicorloris Radix high dose group, middle dose group, low-dose group and Caraganae Erinacea Radix high dose group, middle dose group, low-dose group had highly significant difference compared with model control group (**P<0.01)。...
