Sarcopyramis nepalensis Wall, a natural medicinal plant, is mainly distributed in Fujian, Taiwan, Sichuan, Jiangxi, Hubei, etc. Sarcopyramis nepalensis Wall not only has the efficacies of detoxification, strengthening immunity and resistance hepatitis virus, but also it been used for the treatment of acute or chronic hepatitis in Fujian folk. It is reported that n-butanol part of Sarcopyramis nepalensis Wall is the efficacy of the active site to protect liver. And this part contains saponins. In this thesis, the research focuses on extraction, fractionation and isolation of saponins from Sarcopyramis nepalensis Wall, and it also makes preliminary studies on saponins structure and biological activity.First, organic solvent extraction and system solvent method was employed to extract saponins from Sarcopyramis nepalensis Wall. And the qualitative test of saponins was determined by means of bubble test, acetic anhydride acid solution reaction and dichloroisocyanuric acid reaction. Ginsenoside Rb1 as a standard substance,5% vanillin-glacial acetic acid and perchloric acid as chromogenic reagent, calculate the total saponins content from Sarcopyramis nepalensis Wall. The results showed that the total saponins content from Sarcopyramis nepalensis Wall is 1.8%, the average recovery was 100.65%, RSD is 1.57%.In the next step the inhibition effect of saponins from Sarcopyramis nepalensis Wall was determinated by adopting the method of the filter paper method. Results showed the saponins had stronger inhibition effects on Staphylococcus aureus, Yersinia, Aeromonas hydrophila and Bacillus subtilis. Inhibitory effects on Escherichia coli and Pseudomonas aeruginosa were not obvious.The vitro antioxidant function of saponins from Sarcopyramis nepalensis Wall was determined by the PuLuLan method. The superoxide anion radical scavenging of saponins was observed by ways of pyrogallol oxidation systems, lipid peroxidation by means of TBA. The results showed saponins from Sarcopyramis nepalensis Wall had good antioxidant ability and stronger inhibitory effects on liposome peroxide. It could also scavenge superoxide anion radical.Finally the silica gel column chromatography and preparation of liquid chromatography were suggested to purify saponins.The experiment result showed that the purity of saponins were 92.5%,93.4%,97.8%. The preliminary identification showed that compound Ⅰ and Ⅱ were steroidal saponins.
